60Co-γ射线对小菜蛾遗传不育的影响

摘要:
【目的】采用60Co-γ 200 Gy辐照小菜蛾6日龄蛹,并通过蛹的羽化率及成虫的飞行能力、交配能力、生殖能力、寿命等一系列参数来验证遗传不育技术防治小菜蛾的可行性。【方法】用200 Gy 60Co-γ辐照50头小菜蛾6日龄蛹,然后将其与50头未辐照的对照组6日龄蛹置于温度(25±1) ℃、相对湿度60%~70%、光照时间14~16 h/d、光照强度1 000~4 000 lx的条件下饲养,观察并记录蛹的羽化率、辐照雄虫的飞行能力、雄虫对雌虫性激素的感知能力、雄虫的交配能力及雌、雄成虫的寿命等参数。【结果】经200 Gy 60Co-γ辐照处理后,小菜蛾雄蛹的羽化率为60%,羽化后雄虫寿命为8.83 d;雌蛹羽化率为52.67%,羽化后雌虫寿命为7.90 d。与对照组相比,辐照雄虫的飞行能力、交配能力、对雌虫性信息素的感知能力均无显著差异。4个配对组合UM×UF、IM×UF、UM×IF、IM×IF(其中UF、UM和IF、IM分别表示未辐照和辐照处理的雌虫和雄虫)的当代成虫单雌产卵量分别为23.24,16.34,6.98,5.33粒,F1代卵的孵化率分别为94.83%,75.93%,86.88%和64.15%,F1代化蛹率分别为82.52%,44.11%,21.83%和10.35%;与对照组相比,辐照组当代成虫和F1代单雌产卵量、F1代卵的孵化率和F1、F2代化蛹率均显著降低;辐照组F1代雄虫与雌虫的性别比以组合IM×UF最大,且显著大于对照组。【结论】 用最佳亚不育剂量200 Gy 60Co-γ辐照小菜蛾6日龄蛹,蛹的羽化率、羽化后成虫寿命及生殖力受辐照影响显著降低,辐照雄虫的飞行能力、交配能力及对雌虫性信息素的感知能力不受辐照影响,说明采用辐照不育技术防治小菜蛾是可行的。

Effect of 60Co-γ radiation on inherited sterility of Plutella xylostella (Lepidoptera:Plutellidae)

Abstract:
【Objective】The 6-day-old pupae of diamondback moth were irradiated by 200 Gy 60Co-γ and parameters including eclosion rate,fight ability,mating ability,fertility and longevity were determined to test the technical feasibility of using sterile insect technique to control diamondback moth.【Method】A total of 50 6 day old pupae were irradiated using 200 Gy 60Co-γ before being raised at temperature of (25±1) ℃,relative humidity of 60%-70%,light time of 14-16 h/d,and light intensity of 1 000-4 000 lx.The eclosion rate,fight ability,mating ability,fertility and longevity were determined.【Result】The eclosion rate of male pupae of diamondback moth was 60% and that of female pupae was 52.67%.The longevity of male pupae after emergence was 8.83 d,and that of female was 7.90 d after 200 Gy 60Co-γ irradiation.The male flight ability,mating ability,and perception of female sex hormones were not significantly affected by radiation.The egg amounts of single adult female of combinations UM×UF,IM×UF,UM×IF and IM×IF were 23.24,16.34,6.98 and 5.33.The hatching rates of F1 generation were 94.83%,75.93%,86.88% and 64.15% and the pupation rates were 82.52%, 44.11%, 21.83% and 10.35%,respectively.Compared with the control group,the single female fecundity of F1 generation and pupation rate of F2 generation of irradiation group were significantly reduced.IM×UF had the largest male to female sex ratio of the F1 generation,which was also significantly greater than that of the control group.【Conclusion】 The eclosion rate, reproductive capacity and longevity of Plutella xylostella were reduced by 200 Gy 60Co-γ irradiation on 6-day-old pupae,while the flying ability,mating ability and ability to perceive female sex hormones of male were not affected.Thus,it is feasible to use sterile insect technique to control Plutella xylostella.
