大豆异黄酮对健康大鼠肝脏CB1R、FAAH、DGAT1表达的影响

摘要:
【目的】研究大豆异黄酮(Soy isoflavone,SIF)对大鼠肝脏中大麻素1型受体(CB1R)、脂肪酰胺水解酶(FAAH)和二脂酰甘油酰基转移酶(DGAT1)表达的影响,为SIF活性的研究与应用提供参考。【方法】试验选取40只6周龄SD大鼠,随机分为正常组和低、中、高剂量组,每组10只,分别按0,50,250,500 mg/kg的剂量经口灌胃含SIF的羧甲基纤维素钠溶液4周后,麻醉,采血收集血清,全自动生化仪进行血液生化指标检测。解剖大鼠取肝脏组织制备石蜡切片,HE染色进行病理分析,油红O检测脂滴沉积情况,免疫组织化学、实时荧光定量PCR(以β-actin为内参基因)分别对CB1R、FAAH、 DGAT1蛋白及其基因进行定量分析。【结果】与对照组相比,SIF低剂量组总胆固醇浓度显著下降(P<0.05);中剂量组总胆固醇、甘油三酯和低密度脂蛋白浓度显著下降(P<0.05);高剂量组总胆固醇和低密度脂蛋白浓度极显著下降(P<0.01)。各组大鼠肝脏组织学结构正常,均未见异常病理变化。SIF中、高剂量组肝脏脂滴沉积极显著减少(P<0.01)。与对照组相比,SIF低剂量组大鼠肝脏中FAAH蛋白的表达量显著增多(P<0.05),中、高剂量组CB1R、FAAH和DGAT1蛋白表达量均极显著增多(P<0.01)。与对照组相比,SIF低剂量组大鼠肝脏CB1R、FAAH、DGAT1 mRNA表达量均显著降低(P<0.05),中、高剂量组CB1R mRNA表达量显著增多(P<0.05),高剂量组FAAH mRNA表达量极显著增多(P<0.01)。【结论】SIF可影响肝脏内CB1R、FAAH、DGAT1的表达,且存在明显的剂量差异。

Effects of soy isoflavone on expression of DGAT1,CB1R and FAAH in liver of rats

Abstract:
【Objective】This study investigated the effects of soy isoflavone (SIF) on expression of CB1R,FAAH and DGAT1 in rat liver to provide reference for study and application of SIF.【Method】Forty rats at the age of 6 months were randomly divided into control,low,medium and high dose groups,with ten per group.Sodium carboxy methyl cellulose solutions of SIF at the concentrations of 0,50,250,and 500 mg/kg were given through intragastric administration for 4 weeks.Then,rats were anesthetized and blood serum was collected for biochemical testing with an automatic biochemical analyzer.Rats were also dissected and liver tissues were taken to prepare paraffin sections.HE staining was used to conduct pathological analysis,and oil red O was used for detection of lipid droplet deposition.Immunohistochemistry and real-time quantitative PCR (β-actin as the internal reference gene) were used to quantify protein and gene,respectively.【Result】Compared with the control group,TC content of low dose group decreased significantly (P<0.05).The contents of TC,TG and LDL in the medium dose group were significantly decreased (P<0.05).The contents of TC and LDL in the high dose group were very significantly decreased (P<0.01).The histological structure of liver was normal in rats,and no abnormal pathological changes were observed.Compared with the control group,the deposition of lipid droplets in the medium and high dose groups were significantly reduced(P<0.01).The expression of FAAH was significantly increased (P<0.05),while expression levels of CB1R,FAAH and DGAT1 in the medium and high dose groups were significantly increased (P<0.01).The levels of DGAT1,FAAH and CB1R mRNA in liver of the low dose group were significantly decreased (P<0.05),CB1R mRNA was significantly increased in both medium and high dose groups (P<0.05),and FAAH mRNA in high dose group was significantly increased (P<0.01).【Conclusion】 SIF could affect the expression of CB1R,DGAT1 and FAAH in liver with significant dose difference.
