Rybp基因的表达特性分析

摘要:
【目的】研究牛Rybp基因时空表达图谱,为Rybp基因的功能研究奠定基础。【方法】利用实时定量PCR方法,检测Rybp基因在秦川牛16种组织(心脏、肝脏、脾脏、肺脏、肾脏、睾丸、腹膜脂肪、心包脂肪、背最长肌、里脊、大肠、小肠、瘤胃、网胃、皱胃和瓣胃)、5个不同发育阶段(6,12,18,24和60月龄)肌肉、脂肪组织以及不同分化阶段(第0,2,4,6,8和10天)牛成肌细胞中的表达特性。【结果】Rybp基因在成年秦川牛睾丸组织中的表达量远远高于其他组织,是背最长肌的365倍,而在背最长肌、里脊、心脏等组织中的表达量相对较低。Rybp基因在60月龄肌肉组织中表达量是6月龄的7倍,在6,12,18,24月龄表达量呈微弱上升趋势,但差异不显著;Rybp基因在6,12,18,24月龄脂肪组织中的表达量分别是60月龄的8.5,6.5,5.6和2.1倍,差异达显著水平;Rybp基因在成肌细胞分化第2,4,6,8,10天的表达量分别是第0天的3.6,5.0,9.9,7.0和4.3倍。【结论】随着秦川牛年龄的增加,Rybp基因在肌肉组织中的表达水平逐渐升高,在脂肪组织中的表达水平显著下降;在不同分化阶段成肌细胞中,随着成肌细胞分化进程的推进,Rybp基因表达水平逐渐升高,而当分化减速后,Rybp基因表达水平逐渐降低。牛Rybp基因可能对牛肌细胞分化起抑制作用。

Expression characteristics of cattle Rybp gene

Abstract:
【Objective】The spatio temporal variations of Rybp gene expression in cattle were studied to provide basis for understanding Rybp gene functions.【Method】In this study,real-time quantitative PCR method was used to detect the expression characteristics of Rybp gene in 16 Qinchuan cattle tissues (testis,spleen,lung,liver,retroperitoneal fat,large intestine,kidney,abomasum,rumen,pericardial fat,small intestine,reticulum,omasum,LDM,tenderloin and heart),muscle and fat tissues at 5 different developmental stages (6,12,18,24, and 60 months),and bovine muscle cells at different differentiation stages (0,2,4,6,8,and 10 days).【Result】The expression level of Rybp gene was much higher in testis than in other tissues,whereas the expression levels in LDM,tenderloin and heart were relatively low.The expression in the testicle was 365 times of that in LDM.The expression of Rybp gene in muscle tissue at 60 months was 7 times of that at 6 months,and the expressions in muscle tissue at other months increased slightly without significant difference.The expressions of Rybp gene in fat tissue at 6,8,12 and 24 months were 8.5,6.5,5.6 and 2.1 times of that at 60 months.The expressions of Rybp gene at the 2,4,6,8 and 10 days of muscle cell differentiation were 3.6,5.0,9.9,7.0 and 4.3 times of that at 0 day.【Conclusion】With the growth and development of Qinchuan cattle,the expression level of Rybp gene in muscle tissue increased gradually,while it decreased significantly in adipose tissue.In muscle cells at different differentiation stages,the expression levels gradually increased with the extension of myoblast differentiation,while the expression levels of Rybp gene decreased when the differentiation slowed down.The Rybp gene may inhibit the differentiation of bovine muscle cells.
