柄扁桃根围土AMF的季节变化及与土壤因子的关系

摘要:
【目的】研究柄扁桃根围土丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)的季节动态变化规律以及AMF群落组成与土壤因子之间的相关性,为内蒙古大青山干旱生态系统的保护及应用菌根生物技术培育濒危植物柄扁桃苗木提供理论依据。【方法】以内蒙古大青山干旱阳坡柄扁桃(Amygdalus pedunculata Pall.)根围土为研究对象,采用湿筛倾析 蔗糖离心法分离根围土中的丛枝菌根真菌(AMF)孢子,用形态学方法进行鉴定与分类,并分析AMF群落的多样性,测定土壤全磷、全氮、全钾、有机质、速效磷、碱解氮、速效钾、pH值等指标,然后采用CANOCO 4.5软件对AMF物种与土壤因子之间的相关性进行冗余分析(RDA)。【结果】从柄扁桃不同季节根围土中共分离鉴定出12属38种AMF,其中Funneliformis mosseaeRhizophagus intraradices均为春、夏、秋3个季节共有的优势种。夏季和秋季AMF群落种的丰度和香农 威纳指数均显著高于春季(P<0.05)。春季样本主要受土壤因子全磷(TP)的影响,而夏季和秋季样本主要受土壤因子全氮(TN)的影响。优势种F.mosseaeR.intraradices在春、夏、秋季分别与土壤因子TP、TP和有机质(OM)、速效磷(AP)和OM呈正相关性。【结论】柄扁桃根围土AMF多样性丰富,季节动态变化有明显的规律性,而影响不同季节柄扁桃根围土壤AMF多样性的主要土壤因子有明显差异。

Seasonal changes of AMF in rhizosphere soil of Amygdalus pedunculata Pall and relationships with soil factors

Abstract:
【Objective】This paper investigated the seasonal changes of AMF in Amygdalus pedunculata Pall rhizosphere soil and its correlations with soil factors to provide basis for protecting ecosystem in Daqing Mountains and cultivating Amygdalus pedunculata Pall seedling using mycorrhiza techniques.【Method】The spores of AMF in rhizosphere soil of Amygdalus pedunculata Pall in Inner Mongolia were separated by wet-sieving method and morphological method was applied to classify and identify AMF spores.AMF community structure was analyzed and available nitrogen,available phosphorus,available potassium,organic matter,total nitrogen,total phosphorus,total potassium,and pH were determined by conventional assay methods.Then CANOCO 4.5 software was used to conducted RDA on the relationships between AMF species and environmental factors.【Result】A total of 12 genera and 38 species of AMF were separated from Amygdalus pedunculata Pall rhizosphere soil,and Funneliformis mosseae and Rhizophagus intraradices were dominant in different seasons.Abundance of species and Shannon Weiner index of AMF in summer and fall were higher than those in spring (P<0.05).AMF community was mainly influenced by total phosphorus in spring and by total nitrogen in summer and fall.Dominant species (F.mosseae and R.intraradices) were correlated positively with total phosphorus,total phosphorus and organic matter,and available phosphorus and organic matter in spring,summer,and fall,respectively. 【Conclusion】AMF in the rhizosphere soil of Amygdalus pedunculata Pall was rich with dramatic seasonal variations.Different soil factors affected AMF in different seasons.
