茶多酚对猪精液常温保存效果的影响

摘要:
【目的】 研究茶多酚(TPP)对猪精液常温保存效果的影响。【方法】 手握法采集20月龄健康杜洛克种公猪精液,在基础稀释液中添加不同质量浓度的TPP(0(CK),0.01,0.02,0.04,0.08,0.16 g/L)作为稀释液进行常温(17 ℃)保存,每隔24 h检查精子活率、质膜完整率和顶体完整率,分别在精液保存0,2,4,6 d时检测精液中的细菌含量,并在3 d和6 d时检测精液中的丙二醛含量(MDA)和总抗氧化(T-AOC)活性。【结果】 添加0.02,0.04,0.08 g/L TPP可明显提高猪精子活率、质膜完整率、顶体完整率和T-AOC活性,降低精液的MDA含量和细菌含量。且以0.04 g/L TPP效果最佳,精液有效保存时间为6 d。6 d时0.04 g/L TPP组猪精子活率、质膜完整率、顶体完整率、丙二醛(MDA)含量、总抗氧化(T-AOC)活性和细菌含量分别为60.02%,46.40%,75.25%,2.39 nmol/L,2.31 U/mL和2.19×106 CFU/mL。【结论】 茶多酚能有效提高猪精液常温保存效果,最适添加量为0.04 g/L。

Effects of tea polyphenol on boar sperm quality during room temperature storage

Abstract:
【Objective】 This study aimed to investigate the effects of tea polyphenol (TPP) on room temperature storage of boar sperm.【Method】 Semen samples were collected from 20 months old health Duroc boars with massage method, and diluted with basic liquid containing different concentrations of TPP (0(CK),0.01,0.02,0.04,0.08 and 0.16 g/L). Sperm motility,plasma membrane integrity and acrosome integrity were measured every 24 h,bacterial content were measured at 0,2,4, and 6 d,and malondialdehyde (MDA) content and total antioxidative capacity (T-AOC) were measured at 3 and 6 d during the storage at 17 ℃.【Result】 Basic liquid supplemented with TPP at concentrations of 0.02,0.04,and 0.08 g/L significantly improved sperm motility,plasma membrane integrity,and acrosome integrity obviously,while decreased MDA and bacterial contents in boar semen (P<0.05).The 0.04 g/L TPP treatment had the optimum effect,and the effective storage time of sperm was 6 days.After 6 days,sperm motility,plasma membrane integrity,acrosome integrity, MDA content,and bacterial content were 60.02%,46.40%,75.25%,2.39 nmol/L,2.31 U/mL,and 2.19×106 CFU/mL,respectively.【Conclusion】 TPP could improve quality of boar semen during room temperature storage, and 0.04 g/L TPP was the optimum concentration.
