【Objective】This study researched longhorn beetles and nematode from Japanese five-needle pine (Pinus parviflora) garden to reveal the possibility and spread risk of this pine trees infected by pine wood nematode (Bursaphelenchus xylophilus) and provide theoretical support for quarantine.【Method】The wood and twig samples of dead or dying five needle pine trees were collected from nursery. The woods for inspection of longhorn larvae were split and the longhorn beetle feeding scars on the twigs were examined. Some wood samples were carried to lab by isolating and identifying the nematode and then the longhorn larvae in the wood were further identified under dissect microscope.Other wood samples were kept on a box till the larvae developed into adults and then the adults were captured and dissected under microscope to check whether nematodes were carried.【Result】About 33% of the samples were infected by B.xylophilus;and 33% by B.mucronatus.The larvae were identified as M.alternatus, and the adults after emerging were further identified as the beetle M.alternatus, which carried nematodes LⅣ of B.xylophilus.【Conclusion】The Japanese five needle pine trees could be infested by pine wood nematode B.xylophilus and associated with the vector beetle M.alternatus.The pine trees must be quarantined strictly to avoid dispersal of the pine wilt disease. |
