卵巢摘除对山羊组织IGF-Ⅱ和IGF-ⅡR基因表达量的影响

摘要:
【目的】研究卵巢摘除对布尔山羊杂种母山羊羔肝脏、背最长肌、股二头肌中胰岛素样生长因子Ⅱ(IGF-Ⅱ)及其受体(IGF-ⅡR)基因表达量的变化,从分子水平探讨卵巢摘除对这些组织中IGF-Ⅱ和IGF-ⅡR基因表达量的影响,探讨去卵巢是否可以促进羔羊生长发育。【方法】将体质量相近的5月龄布尔山羊杂种母山羊羔作为研究对象,随机分为处理组和对照组。试验初期摘除处理组母山羊羔卵巢,对照组不摘除。饲养60 d后,取肝脏、背最长肌、股二头肌组织样,运用实时荧光定量PCR法(RT-qPCR)检测其中IGF-Ⅱ和IGF-ⅡR基因mRNA相对表达量。【结果】卵巢摘除后,母山羊羔背最长肌、股二头肌中IGF-Ⅱ基因和肝脏中IGF-ⅡR基因mRNA相对表达量极显著高于对照组(P<0.01),而背最长肌中IGF-ⅡR基因mRNA表达量显著低于对照组(P<0.05),肝脏中IGF-Ⅱ基因和股二头肌中IGF-ⅡR基因mRNA相对表达量低于对照组,但差异不显著(P>0.05)。【结论】摘除卵巢可上调背最长肌和股二头肌中IGF-Ⅱ基因以及肝脏中IGF-ⅡR基因的表达,但下调背最长肌和股二头肌中IGF-ⅡR基因以及肝脏中IGF-Ⅱ基因的表达,表明摘除卵巢有利于母山羊羔肌肉发育和肉质的改进与提高。

Influences of ovariectomy on IGF-Ⅱ and IGF-ⅡR in tissues of Boer goats

Abstract:
【Objective】The objective of this study was to investigate the influences of ovariectomy on relative mRNA expression levels of insulin like growth factor Ⅱ (IGF-Ⅱ) and its receptor (IGF-ⅡR) in hybrid ewes of Boer goats.【Method】The hybrid ewes of Boer goats at the age of about 5 months with similar weights were randomly divided into test and control groups.The ewes in test group were ovariectomized at the beginning and the control group was not.After 60 days,ewes of both groups were selected randomly to slaughter.Real-time quantitative PCR (RT-qPCR) was used to detect the relative mRNA expression levels of IGF-Ⅱ and IGF-ⅡR in liver,longissimus muscle,and biceps femoris. 【Result】The relative mRNA expression level of IGF-Ⅱ gene in longissimus muscle and biceps femoris and that of IGF-ⅡR gene in liver were extremely significantly higher than in control group (P<0.01).The relative mRNA expression levels of IGF-ⅡR gene in longissimus muscle was significantly lower than that in control group (P<0.05),the relative expression level of IGF-Ⅱ gene in liver and that of IGF-ⅡR gene in biceps femoris were lower than those in control group,but the difference was not significant (P>0.05). 【Conclusion】Ovariectomy increased the expression of IGF-Ⅱ in longissimus muscle and biceps femoris and the expression of IGF-ⅡR in liver,but reduced the expression of IGF ⅡR in longissimus muscle and biceps femoris and the expression of IGF-Ⅱ in liver.It is suggested that ovariectomy is advantageous to muscle development of hybrid ewes of Boer goat and improvement and enhancement of meat quality.
