加味四君子汤对仔猪血清免疫因子水平及肠黏膜免疫的影响

摘要:
【目的】探讨加味四君子汤对仔猪免疫功能的影响。【方法】将6窝60头1日龄健康仔猪随机分为3组,每组20头,14日龄开始分别饲喂代乳料、代乳料+1%(以生药计)的加味四君子汤、代乳料+2%(以生药计)的加味四君子汤,21 日龄断奶,试验期14 d。在14日龄、21日龄、28 日龄,每组随机选择10头仔猪,前腔静脉采血,ELISA法检测血清γ干扰素(IFN-γ)、白介素2(IL-2)、IL-4、IL-6水平。在28日龄,各组随机选择6头仔猪剖杀,刮取小肠黏膜,-20 ℃保存,ELISA法检测肠黏膜SIgA水平;取小肠各段分别置于4%多聚甲醛和液氮保存,免疫组化法(IHC)检测SIgA蛋白表达水平,RT qPCR法检测IgA mRNA相对表达量。【结果】加味四君子汤能显著或极显著提高仔猪血清4种免疫因子水平,极显著提高仔猪肠道黏膜SIgA水平(P<0.01),显著或极显著提高小肠SIgA蛋白表达量和IgA mRNA相对表达量,且2%加味四君子汤的添加效果优于1%加味四君子汤。【结论】加味四君子汤能够促进仔猪小肠IgA mRNA表达与SIgA蛋白表达,〖JP〗提高仔猪机体免疫应答能力。

Effect of supplementary Sijunzi decoction on levels of serum immune factors and intestinal mucosa immunity of piglets

Abstract:
【Objective】This experiment was conducted to analyze the effect of supplementary Sijunzi decoction (SJZD) on immune function of piglets.【Method】A total of 60 1-day-old healthy piglets from 6 litters were randomly divided into three groups with 20 piglets each,and fed with milk replacer,milk replacer plus 1%(dry herbs)supplementary Sijunzi decoction,and 2% (dry herbs) supplementary Sijunzi decoction from 14 d on.The trial last for 14 d.Blood samples in ten piglets from each group were taken from vena cava anterior on days 14,21,and 28 to measure serum IFN-γ,IL-2,IL-4,and IL-6 levels by ELISA.At the end of experiment,six piglets in each group were bled to death,and the small intestinal mucosa was collected and stored at -20 ℃ to detect SIgA levels.Each segment of the intestine was also collected and placed in 4% paraformaldehyde and liquid nitrogen to detect expression and distribution of SIgA by IHC and the relative expression of IgA mRNA by RT qPCR.【Result】Supplementary SJZD increased the levels of serum immune factors of piglets significantly or very significantly (P<0.05 or P<0.01),increased the SIgA levels in intestinal mucosa significantly (P<0.01),and elevated protein expression values of SIgA and relative expression values of IgA mRNA in small intestine significantly or very significantly (P<0.05 or P<0.01).The effect of 2% supplementary SJZD was better than that of 1% supplementary SJZD.【Conclusion】Supplementary SJZD could promote the IgA mRNA transcription and SIgA positive expression in small intestine,and enhance immune response of piglets.
