福建NADC30-like PRRSV FJLY01株的全基因组分子特征分析

摘要:
【目的】了解福建省NADC30-like PRRSV FJLY01株的分子生物学特征和遗传演化规律,为PRRSV的防控提供参考。【方法】对采自福建省规模化猪场的疑似病料进行PRRSV分离鉴定,以NADC30 PRRSV毒株基因序列为参考设计并合成9对引物,分别对分离株序列进行分段PCR扩增,经克隆测序后进行序列拼接,采用MEGA 6.0软件进行核苷酸及氨基酸同源性比对,并绘制其与其他35株毒株的系统遗传进化树。【结果】FJLY01株基因组全长为15 016 bp(不包含polyA),包含10个开放阅读框,5′-UTR长190 bp,3′-UTR长148 bp,Nsp2基因缺失393 bp。Nsp2蛋白存在类似PRRSV MN184和NADC30株的131个氨基酸的缺失。核苷酸比对结果表明,FJLY01株与VR-2332、MLV RespPRRS/Repro、CH-1a和BJ-4株核苷酸的同源性为85.2%~86.3%,与JXA1、HuN4等HP PRRSV株核苷酸的同源性为83.7%~84.1%,与MN184A、MN184B株核苷酸的同源性为87.9%~88.2%,与NADC30株核苷酸的同源性最高,为97.1%。系统进化树分析表明,该毒株属于NADC30 like毒株,与JXA1、VR2332和CH-1a的亲缘关系较远。【结论】福建省存在NADC30-like毒株,应该加强PRRSV的监测。

Molecular characterization of NADC30-like PRRSV isolate FJLY01 from Fujian

Abstract:
【Objective】This study investigated molecular epidemiology and genetic characteristics of NADC-30-like porcine reproductive and respiratory syndrome virus (PRRSV) in Fujian to provide reference for the prevention and control of PRRSV.【Method】The sample was collected from a pig farm with clinical signs in Fujian.The virus isolates were propagated in Marc-145 cells.According to the complete genome sequences of NADC30 PRRSV,nine primers were designed and synthesized and the full length genome of FJLY01 was amplified by RT PCR and analyzed by biology information analyzing software.【Result】 The complete genome of FJLY01 was 15 016 bp (excluding polyA),including 10 open reading frames (ORFs) with 190 bp in the 5′-UTR,148 bp in the 3′-UTR,and discontinuous deletion of 393 bp in the Nsp2.Nsp2 was similar to MN184 and NADC30.FJLY01 strain shared 85.2%-86.3% with VR-2332,MLV RespPRRS/Repro,CH-1a and BJ-4,83.7%-84.1% with JXA1 and HuN4,87.9%-88.2% with MN184A and MN184B,and 97.1% with NADC30.The phylogenetic tree analysis showed that FJLY01 isolate was NADC30-like PRRSV.【Conclusion】NADC30-like PRRSV existed in Fujian and it is important to monitor PRRSV evolution in China.
