亚洲棉Argonaute蛋白家族的生物信息学预测与分析

摘要:
【目的】在全基因组水平上鉴定亚洲棉(Gossypium arboretum L.)AGO蛋白家族成员,分析其进化关系,为亚洲棉遗传改良提供基因资源。【方法】用AGO蛋白保守域在亚洲棉基因组数据库中进行同源比对,并对AGO蛋白家族的进化关系、基因结构、结构域、染色体定位、三级结构及表达模式等进行生物信息学分析。【结果】从亚洲棉蛋白数据库中共鉴定出11个AGO蛋白家族成员,其编码区长度为2 949~9 807 bp,定位在6条染色体上,其中第9号染色体上分布有4个基因;11个AGO家族成员分为5个亚族,亚族内各成员间具有很高的保守性;AGO家族成员均具有典型的α-螺旋跨膜结构,属于碱性蛋白;从表达模式看,AGO家族成员在纤维发育不同时期和不同组织中的表达量存在较大差异,其中5个基因在棉纤维次生壁后期高表达,在其他时期表达水平较低,存在组织特异性。【结论】成功鉴定了亚洲棉AGO蛋白家族,明确了其基本结构的进化关系及基因表达特点。

Bioinformatics analysis of Argonaute protein family in Gossypium arboretum

Abstract:
【Objective】This study identified Argonaute (AGO) proteins from Gossypium arboretum at the whole genome level and analyzed the evolutionary relationship to lay foundation for genetic engineering improvement of G.arboretum.【Method】AGO protein conserved domain was compared with those in Asian cotton genome database,and bioinformatics method was used to investigate the relationship between the evolution,gene structure,domain structure,chromosomal location,tertiary structures and expression patterns.【Result】A total of 11 AGO members with length of 2 949 to 9 807 bp were identified,and they were located in 6 chromosomes with 4 in the ninth chromosome.The proteins can be divided into five subfamilies and they were highly conserved.All family members had α-helix and were basic protein.Members of the family expressed differently in different development periods.Five genes were highly expressed in the secondary wall of the cotton fiber,but were lowly expressed in other periods,showing tissue specificity.【Conclusion】The identification and bioinformatics analysis provided support for future study of AGO genes in G.arboretum.
