响应面法优化发芽苦荞富集γ-氨基丁酸的培养条件

摘要:
【目的】研究发芽苦荞富集γ-氨基丁酸(γ-aminobutyric acid,GABA)的最佳培养条件,为苦荞芽菜的开发利用提供支持。【方法】在谷氨酸钠(Monosodium glutamate,MSG)质量浓度、钙离子(Ca2+)浓度、发芽温度和发芽时间4个单因素试验基础上,采用响应面法对发芽苦荞的培养条件进行优化,建立GABA富集的二次多项式数学模型,并分析模型的有效性与因素间的交互作用。【结果】4个因素对GABA富集的影响大小依次为MSG质量浓度>发芽温度>发芽时间>Ca2+浓度;发芽苦荞富集GABA的最佳培养条件为:MSG浓度6.90 mg/mL,发芽温度32 ℃,发芽时间3.5 d,Ca2+浓度5.8 mmol/L,在此条件下发芽苦荞GABA含量为(258.77±3.95) μg/g,与预测值259.89 μg/g相差较小。【结论】建立的回归模型能够较准确地预测发芽苦荞的GABA富集量。该培养方法稳定性好,GABA富集量高。

Optimizing culture conditions for GABA accumulation in germinated tartary buckwheat based on response surface methodology

Abstract:
【Objective】The optimal culture conditions for γ-aminobutyric acid (GABA) accumulation in germinated tartary buckwheat were obtained to provide support for the development and utilization of tartary buckwheat sprouts.【Method】On the basis of four single factor tests,response surface methodology was adopted to optimize the germinated conditions including monosodium glutamate (MSG) mass concentration,calcium ion (Ca2+) concentration,temperature,and time.A second order quadratic equation was established and the applicability of the model and interactions between factors were analyzed.【Result】The effects of the four factors on GABA accumulation were in a decreasing order of MSG mass concentration>temperature>time>Ca2+ concentration.The optimal conditions for GABA accumulation were MSG concentration of 6.90 mg/mL,temperature of 32 ℃,time of 3.5 d and Ca2+ concentration of 5.8 mmol/L.The accumulation of GABA was (258.77±3.95) μg/g under the optimal conditions,which was similar to the predicted value of 259.89 μg/g.【Conclusion】The established model well predicted the accumulation of GABA and the optimal culture method was stable with high GABA accumulation.
