‘美人指’葡萄果实糖积累和蔗糖代谢相关酶活性的研究

摘要:
【目的】研究避雨栽培模式下‘美人指’葡萄果实的生长发育、糖分积累以及蔗糖代谢相关酶活性,探讨果实发育过程中糖分积累与酶活性的关系,为避雨条件下葡萄的优质栽培提供理论依据。【方法】以“地膜+天膜”避雨栽培下5年生‘美人指’葡萄为供试材料,于谢花后20 d开始每10 d取样1次,测定葡萄果实的纵径、横径、单果质量和果糖、葡萄糖、蔗糖含量以及4种蔗糖代谢相关酶活性的动态变化。【结果】‘美人指’葡萄果实呈双“S”形曲线生长模式,并存在2个快速生长阶段;果实糖分积累主要以葡萄糖和果糖为主,葡萄糖含量一直明显高于果糖;在果实整个发育期,酸性转化酶和蔗糖合成酶分解方向的活性一直较高,蔗糖磷酸合成酶活性很低;且蔗糖含量与蔗糖磷酸合成酶活性呈显著正相关,与酸性转化酶活性呈显著负相关。【结论】蔗糖分解方向的酶活性远大于合成酶类,促进了葡萄糖和果糖的积累,且酸性转化酶和蔗糖磷酸合成酶是‘美人指’葡萄果实糖分积累的关键酶。

Abstract:
【Objective】The research investigated the evolution of sugar accumulation and sucrose metabolism related enzymes activities of ‘Manicure Finger’ grape during the growth process under rain-shelter cultivation and discovered the relationship between sugar accumulation and metabolizing enzymes in grapes to provide theoretical support for cultivating high quality grapes.【Method】Five years old ‘Manicure Finger’ vines under rain shelter cultivation were selected to measure the longitudinal diameter, transverse diameter,fruit weight,soluble sugar content,and activities of sucrose metabolism-related enzymes every 10 days.【Result】The growth of ‘Manicure Finger’ fruits followed a double “S” growing curve.There were two rapid growth stages.The sugar of fruits mainly accumulated with the forms of glucose and fructose,and the content of glucose was always higher than that of fructose.During the entire growth process,both acid invertase activity and sucrose synthase activity were high in cleavage direction,while the activity of sucrose phosphate synthase was very low.The sucrose content had significantly positive relationship with sucrose phosphate synthase activity,and significantly negative correlation with acid invertase activity.【Conclusions】Both acid invertase and sucrose phosphate synthase played dominate roles in sugar accumulation.The sucrose-metabolizing enzymes had much higher activities than the sucrose synthesis enzymes,which promoted the accumulation of glucose and fructose.
