西瓜杂交组合T-1种子纯度鉴定的SSR标记研究

摘要:
【目的】研究有效的西瓜杂交组合种子纯度的SSR标记鉴定方法,为室内鉴定的实际应用提供理论依据。【方法】以西瓜杂交组合T-1及其亲本自交系P-1、M-13为材料,在子叶展平期采用CTAB法提取DNA,利用优化的西瓜SSR标记方法进行杂交种子的纯度鉴定,并与田间表型鉴定进行比较。【结果】从36对SSR引物中筛选出3对(MCPI-5、MCPI-16、MCPI-17)稳定性强、可重复性好、在杂交种及其母本之间具有多态性的引物。将这3对引物中的MCPI-5和MCPI-16用于267株T-1杂交种群体纯度鉴定,显示种子纯度为97.75%,与田间种植鉴定结果一致。【结论】3对SSR引物MCPI-5、MCPI-16和MCPI-17可用于西瓜杂交组合T-1种子纯度的室内鉴定。

Purity identification of watermelon hybrid combination T-1 seeds using SSR molecular marker

Abstract:
【Objective】SSR molecular marker technique was used to identify purity of watermelon hybrid seeds,aiming to provide a reliable indoor method for purity identification of hybrid seeds.【Method】Seeds of watermelon hybrid combination T-1 and its parents (P-1,M-13) were selected in this study.DNA was extracted with CTAB method in cotyledon and optimized with SSR molecular marker method.SSR identification results were compared with field identification.【Result】Three pairs of primers,which were stability,repeatability and polymorphic between hybrids and their parents,were selected from 36 pairs of SSR primers.A total of 267 seedlings of T-1 were identified by MCPI-5 and MCPI-16,the obtained purity was 97.75%,identical to field identification.【Conclusion】Three pairs of SSR primers (MCPI-5,MCPI-16 and MCPI-17) could be used to identify the seeds purity of T-1.
