忍冬开花过程中花青苷组成及含量的变化

摘要:
【目的】探讨忍冬属(Lonicera Linn.)植物花朵中花青苷的种类及开花过程中含量的变化,为忍冬花色育种提供依据。【方法】以忍冬(L.japonica Thunb.)、‘火焰’忍冬(L.×heckrottii Rehd.‘Huoyan’)、‘格雷姆’忍冬(L.periclymenum L.‘Geleimu’)、贯月忍冬(L.sempervirens L.) 和‘台尔曼’忍冬(L.×tellmanniana)5种藤本植物为材料,采用高效液相色谱-二极管阵列检测技术(HPLC-DAD)、高效液相色谱-电喷雾离子化-质谱联用技术(HPLC-ESI-MSn),对各材料开花过程中的花青苷进行定性及定量分析。【结果】在开花过程中,5种忍冬属植物花色变化明显,但在忍冬花朵中未检测到花青苷存在,在‘火焰’忍冬、贯月忍冬、‘格雷姆’忍冬和‘台尔曼’忍冬中共检测到4种花青苷,分别为矢车菊素3,5-二葡萄糖苷、矢车菊素3-葡萄糖苷、芍药花素3,5-二葡萄糖苷和芍药花素3-葡萄糖苷,其中矢车菊素3,5-二葡萄糖苷为主要花色素成分。5种忍冬属植物在不同开花阶段花青苷种类基本一致,但含量均有较为明显的变化。【结论】5种忍冬属植物的花青苷组成及含量变化与其花色转变相一致,可为忍冬花色育种提供参考。

Changes in composition and content of anthocyanins during flowering process of Lonicera

Abstract:
【Objective】This study investigated the changes in anthocyanins composition and content in Lonicera during flowering to provide theoretical basis for development and breeding of flower colors.【Method】A high performance liquid chromatography (HPLC) with a photodiode array detector (HPLC-DAD) and a HPLC-electrospray ionization-multiple mass spectrometry (HPLC-ESI-MSn) were employed for qualitative and quantitative analysis of anthocyanins from L.japonica Thunb.,L.×heckrottii Rehd.‘Huoyan’,L.periclymenum L.‘Geleimu’,L.sempervirens L.and L.×tellmanniana.【Result】During flowering process,the colors of all varieties changed significantly.But no anthocyanins were detected in flowers of L.japonica during flowering process.In L.×heckrottii Rehd.‘Huoyan’,L.sempervirens L.,L.periclymenum L.‘Geleimu’ and L.×tellmanniana,a total of 4 anthocyanins including Cy 3,5-di-O-glucoside,Cy 3-O-glucoside,Pn 3,5-di-O-glucoside,and Pn 3-O-hexoside were detected.Cy 3,5-di-O-glucoside was the main pigment.The types of anthocyanins from different varieties were almost same but their contents were significantly different.【Conclusion】The transformation of flower colors was consistent with composition and content of anthocyanins in Lonicera flower.This study could provide reference for breeding and selection of special flower colors.
