催产素和苯甲酸雌二醇对小尾寒羊外周血淋巴细胞增殖的影响

摘要:
【目的】研究催产素(OXT)和苯甲酸雌二醇(E2)对淋巴细胞增殖的影响,探讨OXT和E2的相互作用机制。【方法】分别以静息和Con A刺激下的雌性绵羊外周血淋巴细胞为研究对象,用不同质量浓度(低质量浓度(OXT 10 ng/mL,E2 50 pg/mL)、中质量浓度(OXT 100 ng/mL,E2 500 pg/mL)和高质量浓度(OXT 1 000 ng/mL,E2 5 000 pg/mL))和处理方式(单用、联用(二者同时加入、E2预处理12 h后加OXT、OXT预处理12 h后加E2))的OXT、E2处理细胞,用MTT试验检测淋巴细胞的增殖水平,评估雌激素、催产素的相互作用方式。【结果】OXT对活化和静息状态的淋巴细胞均有抑制作用,且呈剂量依赖性。低质量浓度的E2可促进Con A刺激的T淋巴细胞增殖,但对静息状态的淋巴细胞无明显影响;随E2质量浓度的增加,高质量浓度的E2对不同状态的淋巴细胞增殖均有抑制作用。低质量浓度OXT和E2联用条件下,Con A活化的T淋巴细胞的刺激指数(SI)值介于2种激素单独作用之间;静息状态下的淋巴细胞增殖明显受到抑制,该抑制作用均强于2种激素单独作用;用生理浓度的OXT(10 ng/mL)和E2(50 pg/mL)对活化状态的淋巴细胞分别进行先后处理,对不同状态淋巴细胞增殖的影响偏向于后加入的激素的作用。【结论】高质量浓度的E2可抑制静息和Con A刺激活化的淋巴细胞的增殖,低质量浓度的E2对Con A刺激活化的淋巴细胞增殖有促进作用,而OXT抑制淋巴细胞的增殖。高质量浓度OXT和E2联用,在静息状态的淋巴细胞上体现了协同作用;低质量浓度OXT和E2联用,在活化状态的淋巴细胞上体现了相互颉颃和“利他作用”2种作用模式,显示了二者相互作用方式的多样性。

Effects of oxytocin and estradiol benzoate on proliferation of peripheral blood lymphocytes of Small-tail Han sheep in vitro

Abstract:
【Objective】This study investigated the effects of oxytocin (OXT) and estradiol (E2) on proliferation of peripheral blood lymphocytes and explored the interaction between OXT and E2.【Method】The peripheral blood lymphocytes of female sheep at rest and stimulated by Concanavalin A (Con A) were treated by different concentrations of OXT and E2 (low concentrations of OXT 10 ng/mL and E2 50 pg/mL,medium concentrations of OXT 100 ng/mL and E2 500 pg/mL,and high concentrations of OXT 1 000 ng/mL and E2 5 000 pg/mL) and by different treatment methods (single use and combined use: simultaneous addition,E2 was 12 h after OXT,and OXT was 12 h after E2) before MTT assay was used to test the proliferation of lymphocytes and interaction of OXT and E2 was evaluated.【Result】OXT had suppressive effect on both active and rested lymphocyte at low dose.E2 at low concentrations could promote the proliferation of T lymphocytes stimulated with Con A but had no effects on lymphocytes at rest.As the increase of concentration,E2 had suppressive effect on lymphocytes at different statuses.Thus,for the combination of OXT and E2 at low concentrations,the stimulation index (SI) of T lymphocytes stimulated with Con A was between the SI values of OXT or E2 alone.The lymphocytes at rest were significantly suppressed and the suppressive effect was stronger than OXT or E2 alone.Active lymphocytes treated with OXT and E2 at normal concentrations in succession had similar effects on proliferation of lymphocytes to adding hormone later.【Conclusion】E2 at high concentration inhibited the proliferation of lymphocytes,whereas it promoted active lymphocytes at low concentration.OXT inhibited proliferation of lymphocytes.OXT and E2 at high concentration had synergy effect on lymphocytes at rest.OXT and E2 at low concentration had antagonism and altruistic effects on lymphocytes stimulated by Con A.
