苏尼特家养双峰驼mtDNA Cyt b基因和D-loop序列的遗传多样性

摘要:
【目的】研究苏尼特家养双峰驼的遗传多样性,以全面揭示中国双峰驼的起源进化,为科学保护和利用我国的双峰驼遗传资源奠定基础。【方法】采集15峰苏尼特家养双峰驼(10峰雄驼和5峰雌驼)的血样,提取其DNA,采用PCR方法扩增线粒体DNA(mtDNA)细胞色素b (Cyt b)基因的全序列及D-loop的671 bp序列,进行遗传多样性分析,并结合GenBank中已有的双峰驼的mtDNA Cyt b基因序列和D-loop序列进行系统发育分析。【结果】苏尼特家养双峰驼mtDNA Cyt b基因中存在10个变异位点,共形成了7种单倍型,单倍型多样度为0.857±0.065,核苷酸多样度为0.001 94±0.002 70,平均核苷酸差异数为2.210;在D-loop的671 bp序列中,存在6个变异位点,共定义了6种单倍型,单倍型多样度为0.714±0.116,核苷酸多样度为0.002 53±0.002 75,平均核苷酸差异数为 1.695。基于Cyt b基因和D-loop序列进行的系统发育分析均显示,苏尼特家养双峰驼与野生双峰驼归于2个不同的母系世系,野生双峰驼不是苏尼特家养双峰驼的直接祖先。【结论】苏尼特家养双峰驼mtDNA Cyt b基因和D-loop序列遗传多态性均比较丰富;苏尼特家养双峰驼与野生双峰驼归于2个不同的母系世系,野生双峰驼不是苏尼特家养双峰驼的直接祖先;家养双峰驼内部存在着一定的遗传分化。

Abstract:
【Objective】The paper studied the genetic diversity of Sunite domestic bactrian camel to reveal the origin and evolution of bactrian camel in China and improve the protection and utilization of bactrian camel genetic resources.【Method】The mtDNA Cyt b gene and D-loop of 15 Sunite domestic bactrian camels (10 male and 5 female) were sequenced and whole length mitochondrial DNA (mtDNA) cytochrome b (Cyt b) gene and D-loop 671 bp sequence were obtained using PCR.Then genetic diversity and phylogenetic analysis were conducted based on obtained sequences and known sequences from GenBank.【Result】There were 10 mutation sites and 7 different haplotypes in Cyt b gene of Sunite domestic bactrian camels,and their haplotype diversity,nucleotide diversity and average number of nucleotide differences were 0.857±0.065,0.001 94±0.002 70,and 2.210,respectively.In the 671 bp D-loop sequences,there were 6 mutation sites and 6 different haplotypes,and their haplotype diversity,nucleotide diversity and average number of nucleotide differences were 0.714±0.116,0.002 53±0.002 75,and 1.695,respectively.Neighbor joining (NJ) tree of mtDNA Cyt b gene and D-loop sequence showed that Sunite domestic bactrian camel and wild bactrian camel belonged to two different lineages and wild bactrian camel was not the origin of domestic bactrian camel.【Conclusion】Genetic diversity in mtDNA Cyt b gene and D-loop of Sunite domestic bactrian camel were rich.Sunite domestic bactrian camel and wild bactrian camel belonged to two different lineages,and wild bactrian camel was not the origin of domestic bactrian camel.There was genetic differentiation in domestic bactrian camels.
