羊口疮病毒新疆流行株的分离鉴定及其遗传进化分析

摘要:
【目的】研究羊口疮病毒(Orf virus,ORFV)新疆流行株的遗传变异情况。【方法】采集新疆石河子地区疑似感染ORFV的羔羊唇部结痂病料,通过PCR扩增、Vero传代细胞分离培养、电镜观察、动物回归试验等方法,检测、分离和鉴定ORFV毒株,对分离鉴定的3株ORFV毒株保护性抗原基因B2LF1L及毒力基因VIRGIFVEGF分别进行克隆、测序及遗传进化分析。【结果】从病料中分离鉴定出3株ORFV新疆流行株(ORFV-SHZ1、ORFV-SHZ2和ORFV-SHZ3)。分析结果显示,ORFV分离株保护性抗原基因B2LF1L相对保守,B2LF1L基因编码氨基酸序列与其他参考株的同源性分别为88.6%~97.9%和86.7%~97.4%;3个毒力基因的变异相对较大,尤其是VEGF基因。VIRGIFVEGF基因编码氨基酸序列与其他参考株的同源性分别为91.8%~97.3%,85.2%~97.0%和71.5%~98.5%。基于B2LVIR基因的遗传进化树分析表明,ORFV-SHZ1和ORFV-SHZ2分离株均与台湾分离株的亲缘关系最近。【结论】试验分离的ORFV新疆流行株可能是由台湾株与其他毒株重组后产生的,且其毒力基因发生了较大的变异。

Isolation,identification and phylogenetic analysis of Orf virus epidemic strains in Xinjiang

Abstract:
【Objective】This study aimed to investigate the genetic evolution situation of Orf virus (ORFV) epidemic strains in Xinjiang.【Method】Samples were collected from lamb with suspected ORFV infection in Shihezi region,and the strains were identified by PCR amplification,isolation and culture in Vero cell,electron microscopy,and animal regression.The major protective antigen genes B2L and F1L and virulence genes VIR,GIF and VEGF of the three identified ORFV were cloned,sequenced,and phylogenetically analyzed.【Result】Three ORFV isolates(ORFV-SHZ1,ORFV-SHZ2,and ORFV-SHZ3) were isolated and identified from collected samples.ORFV protective antigen genes B2L and F1L were relatively conservative,sharing 88.6%-97.9% and 86.7%-97.4% identities in amino acid sequence with other reference strains.The virulence gene variation was significant,especially for VEGF gene.Amino acid identities of VIR,GIF and VEGF genes with other reference strains were 91.8%-97.3%,85.2%-97.0%,and 71.5%-98.5%,respectively.Phylogenetic tree analysis based on B2L and VIR genes showed that ORFV-SHZ1 and ORFV-SHZ2 strains were close to isolates from Taiwan.【Conclusion】Recombination occurred among Shihezi isolates,Taiwan strains and other strains,causing great variation in virulence genes.
