CaCl2对低钙土壤中可培养放线菌数量及种类的影响

摘要:
【目的】分析CaCl2对碳酸钙含量较低土壤中可培养放线菌数量及种类的影响,探索建立新的放线菌分离方法。【方法】以海拔高度及碳酸钙含量不同的秦岭太白山北坡山地土壤为材料,通过稀释平皿涂抹法分离放线菌,研究了高氏1号培养基中加入CaCl2后低钙土壤中可培养放线菌数量及种类的变化,并用菌落形态特征和16S rDNA序列鉴定新出现的放线菌。【结果】向高氏1号培养基中加入CaCl2后,供试低钙土壤样品中可培养放线菌总数及链霉菌数量减少,同时所有供试土壤样品中均有部分在高氏1号培养基上生长的放线菌种类消失;供试土壤中有90种新出现的放线菌种类,从中获得了29株代表性菌株。根据菌落形态特征及16S rDNA序列鉴定可知,新出现的29株放线菌主要为链霉菌属和小单孢菌属,其中65.5%具有生物活性及其他应用价值。【结论】向高氏1号培养基中加入CaCl2,可从低钙土壤中分离到一些新的放线菌,其中有较多的活性物质产生菌。

Abstract:
【Objective】This study aimed to analyze the effects of CaCl2 on number and type of culturable actinomycete in low CaCO3 soils and develop a new isolation method for actinomycete.【Method】Soil samples with different CaCO3 contents were collected at different elevations from Taibai Mountain,Shaanxi Providence,China.The change in number and type of culturable actinomycete after adding CaCl2 to Gause’s synthetic agar in low calcium soils was analyzed by isolating actinomycetes using soil dilution plate technique.The isolates cultured only on medium plus CaCl2 were identified based on 16S rDNA sequence analysis.【Result】Cultured actinomycete and streptomycete counts decreased and some actinomycete species disappeared in CaCl2 treatment in all tested samples.A total of 90 actinomycete isolates were only isolated on Gause’s synthetic agar plus CaCl2.Based on the morphological characters and 16S rDNA sequence analysis,29 typical isolates were selected and they were mainly Streptomyces spp. and Micromonospora spp.,of which 65.5% showed bioactivities or other application values. 【Conclusion】Supplement of CaCl2 to Gause’s synthetic agar could isolate actinomycete species,including many types that could produce bioactive compounds.
