四川黑茶的高效液相色谱指纹图谱研究

摘要:
【目的】应用高效液相色谱法(HPLC)研究四川黑茶的指纹图谱,为四川黑茶的鉴别和质量评价提供参考。【方法】以四川省11个不同生产厂家的22个黑茶样品及我国其他黑茶主产区的13个黑茶样品为试验材料,采用线性梯度洗脱,以甲醇和体积分数2%乙酸水溶液为流动相,在柱温30 ℃,检测波长280 nm,流速1.0 mL/min条件下构建四川黑茶HPLC指纹图谱的共有模式,分析各样品与该共有模式的相似度,并对35个黑茶样品进行聚类分析。【结果】确定了14个色谱峰为四川黑茶的共有峰;精密度、重现性、稳定性试验中各共有峰的相对峰面积、相对保留时间的相对标准偏差(RSD)均小于3.00%;22个四川黑茶样品与共有模式的相似度为0.825~0.978;对相对峰面积的聚类分析结果表明,当临界值为10时,35个来自全国不同产地的黑茶样品可聚为4类,来自四川的22个黑茶样品可聚为一大类。【结论】根据构建的四川黑茶的HPLC指纹图谱,可以鉴定和评价除方包茶之外的四川黑茶。

Abstract:
【Objective】The fingerprinting of Sichuan dark tea was studied using HPLC to improve its identification and quality assessment.【Method】22 Sichuan dark tea samples from 11 different manufacturers and 13 dark tea samples from other regions of China were selected as experimental materials.A suitable chromatographic system was established using a linear gradient elution with methanol and water containing 2% acetic acid as the mobile phase,a detection wavelength of 280 nm and the flow speed of 1.0 mL/min.The common pattern of HPLC fingerprinting of Sichuan dark tea was established,and the similarity index between dark tea samples and the common pattern of Sichuan dark tea were analyzed.The methodology was inspected and cluster analysis of the 35 dark tea samples was conducted.【Result】There were 14 peaks in chromatograms were determined.The RSD of the relative peak area and the relative retention time of common peaks in test of precision,repeatability,and stability were all less than 3.00%.The similarity index between 22 Sichuan dark tea samples and the common pattern of Sichuan dark tea ranged from 0.825 to 0.978.The cluster analysis on the relative peak area showed that the 35 dark tea samples from different origins could be divided into four types,and the 22 Sichuan dark tea samples belonged one same category.【Conclusion】The established HPLC fingerprinting can be used to identify and evaluate Sichuan dark teas except Fangbao tea.
