加味二术散对断奶仔猪肠道菌群的影响

摘要:
【目的】研究加味二术散对断奶仔猪回肠、盲肠肠道菌群的影响,为其在生产中的应用提供依据。【方法】选用30日龄断奶仔猪75头,随机分为5个组,即:不做任何处理的空白对照组,饲喂在基础日粮中添加 0.2%,0.3%和0.5%加味二术散 的3个中药处理组(Ⅰ、Ⅱ、Ⅲ)以及饲喂在基础日粮中添加0.5%硫酸粘杆菌素的阳性对照组,每处理3个重复,每重复5头猪。在饲养第1,2,3周无菌采集空白对照组、中药添加Ⅱ组和阳性对照组仔猪回肠、盲肠内容物,采用PCR DGGE技术分析肠道细菌菌群的结构变化。对DGGE图谱中的差异性和共性条带进行克隆、测序和序列分析,对其对应的菌种进行鉴定。【结果】中药添加Ⅱ组和阳性对照组仔猪肠道菌群的多样性高于空白对照组;盲肠菌群多样性高于回肠。中药添加Ⅱ组和阳性对照组回肠微生物菌群相似性高,均达到82%以上;中药添加Ⅱ组盲肠微生物菌群相似性高于空白对照组和阳性对照组。DGGE图谱中特异条带和共性条带测序结果表明,断奶仔猪回肠、盲肠的肠道细菌主要归属于厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)、放线菌门(Actinobacteria)和变形菌门(Proteobacteria)。【结论】加味二术散能够增加仔猪肠道微生物菌群多样性,改善肠道微生物菌群结构,促进断奶仔猪肠道微生物区系达到新的平衡,防治仔猪腹泻。

Effects of Two Macrocephala Flavored Powder on intestinal microbial populations of newly weaned pigs

Abstract:
【Objective】Effects of Two Macrocephala Flavored Powder (TMFP) on intestinal microbial populations in ileum and cecal of newly weaned pigs were studied.【Method】A total of 75 30-day-old newly weaned pigs were randomly divided into five groups:Normal control group (treated with basic diets),three TMFP groups with concentrations of 0.2%,0.3%,and 0.5% (Ⅰ,Ⅱ,and Ⅲ), and positive control group (treated with antibiotic),each with three repeat,repeat every five pig.Contents of the ileum and cecum of normal control group,TMFP group and positive group were sampled aseptically,PCR-DGGE technology was employed to analyze the bacterial community structures of each group at the end of the first,second,and third weeks.The specific and commonness bands of DGGE were chose to clone,sequencing,analysis and identification.【Result】The bacteria diversity of TMFP Ⅱ group and antibiotic dose group was higher than that of control group,and the bacteria diversity of cecum was higher than that of ileum.The bacterial populations in ileum of TMFP Ⅱ group and antibiotic group had high similarities (>82%).The similarity of bacterial populations in cecum of TMFP Ⅱ group was higher than those of control group and antibiotic group.According to the specific and commonness bands of DGGE,the bacterial in ileum and cecal of newly weaning pigs were mainly from Firmicutes,Bacteroidetes,Actinobacteria and Proteobacteria.【Conclusion】TMFP could increase gastrointestinal microbial diversity,change the structure of intestinal microbial populations,promote intestinal microbial populations to new balance,and prevent the diarrhea.
