长白山区不同花色杓兰属植物遗传多样性的ISSR分析

摘要:
【目的】分析长白山地区不同花色杓兰属植物的遗传多样性。【方法】采用ISSR分子标记技术,对长白山区8个花色11个杓兰属植物个体的遗传多样性进行分析,用Popgen32软件分析Nei’s基因多样性和Shannon信息指数,采用UPGMA法进行聚类分析。【结果】筛选出11条ISSR引物,共扩增出94条条带,其中多态性条带86条,多态基因位点比例为91.5%,平均有效等位基因数为1.418 2,平均Nei’s基因多样性为0.250 0,平均Shannon信息指数为0.369 7,样品间的遗传距离为0.010 5~0.969 7。【结论】长白山区杓兰属植物具有较丰富的遗传多样性,聚类结果与形态学分类结果基本一致。

ISSR analysis for genetic diversity of Cypripedium in Changbai Mountain area

Abstract:
【Objective】The objective of this article was to analyze the genetic diversity of Cypripedium plants with different colors.【Method】The genetic diversity of 11 Cypripedium individuals belonging to 8 color types in Changbai Mountain area was analyzed using ISSR marker.Nei’s genetic diversity index and Shannon information index were calculated using Popgen 32 software.Cluster analysis was also conducted using UPGMA.【Result】Eleven primers were selected and applied for PCR amplification.A total of 94 bands were produced and 86 of them were polymorphic with the PPB of 91.5%.The average effective number of alleles,Nei’s gene diversity and Shannon information index were 1.418 2,0.250 0,and 0.369 7,respectively.The distance between 11 individuals was 0.010 5-0.969 7.【Conclusion】Cypripedium plants in Changbai Mountain area had comparatively great genetic diversity.Clustering analysis results were generally consistent with morphological taxonomy results.
