镉对生长期小鼠脾组织及脾细胞DNA的损伤作用

摘要:
【目的】研究镉对生长发育阶段小鼠脾组织及脾细胞DNA的损伤作用,以揭示镉的免疫毒性作用机制。【方法】将生长期小鼠随机分为对照组、镉低剂量组(LD50/100,经口灌胃测定镉的半数致死剂量LD50为187 mg/kg)、镉中剂量组(LD50/50)和镉高剂量组(LD50/25),饮水染镉40 d时处死,分析脾脏相关参数和血液免疫指标变化,利用光学显微镜检查脾脏组织的病理学变化,利用彗星试验研究脾细胞DNA的损伤情况。【结果】随着染镉剂量的增加,小鼠的脾脑系数和脾脏指数均增大;白细胞总数和淋巴细胞比率升高,中性粒细胞比率不变,中值细胞比率、免疫球蛋白G和免疫球蛋白M降低;各镉剂量组小鼠脾脏组织的形态结构均遭到损伤,镉高剂量组最严重,表现为白髓区淋巴细胞数量明显减少,红髓内红细胞数量明显增加;各镉剂量组小鼠脾细胞DNA 的拖尾率、尾长、彗星全长、尾矩、Olive 尾矩和损伤等级均高于对照组,呈明显的剂量 效应关系。【结论】镉可致生长期小鼠的脾脏相关参数、血液免疫指标、组织形态结构改变和脾细胞DNA损伤,从而对其免疫功能产生抑制性影响。

Abstract:
【Objective】This research was conducted to study the damage of cadmium to spleen tissues and splenocytes DNA of growing mice.【Method】Growing mice were randomly divided into four groups,control group,low dose group (LD50/100),middle dose group (LD50/50) and high dose group (LD50/25).LD50,the median lethal dose of cadmium chloride via oral administration to mice,was 187 mg/kg BW.Mice were sacrificed after freely drinking water containing cadmium chloride or deionized water for 40 days,consecutively.The changes of spleen relevant parameters and hematogenic immunity indexes were analyzed,splenic histopathological changes were observed by microscope,and splenocytes were used to investigate DNA damage by comet assay.【Result】With the increase of cadmium concentration in growing mice,spleen/brain weight ratio and spleen/body weight ratio increased.Total white blood cells and lymphocytes ratios rose,and neutrophilic granulocyte ratios did not change,but mid value cells ratios,IgG and IgM all reduced.The splenic morphosis was destroyed and become more and more serious with the increase of cadmium exposure dose,and the most serious disorganization was in the high dose group,in which lymphocytes decreased in white pulp area and red blood cells increased in red pulp area.The ratio of cells tailing,DNA tail length,comet length,tail moment,olive tail moment and percentage of DNA damage in every cadmium exposure group were all higher than that of the control group in a dose-response relationship.【Conclusion】Cadmium could induce the change of spleen relevant parameters,hematogenic immunity indexes,splenic morphosis and damage to splenocytes DNA of growing mice,so as to inhibit the immune function of mice.
