【目的】优化脂肪酶催化合成共轭亚油酸植物甾醇酯的工艺条件。【方法】在水溶剂体系中以脂肪酶Pseudomonas为催化剂,研究了共轭亚油酸植物甾醇酯的合成工艺条件,并对产物进行了气相色谱分析与测定;在单因素试验的基础上,选取反应时间、酶添加量、反应温度作为影响酯化反应的主要因素,以酯化率为响应值进行响应面分析,构建相应的回归模型并进行验证。【结果】共轭亚油酸植物甾醇酯的最佳合成工艺条件为:反应温度55.5 ℃、酶添加量6.6%、反应时间41.2 h,在此条件下,酯化率达到84.37%。【结论】脂肪酶催化共轭亚油酸植物甾醇酯合成工艺的研究为甾醇酯安全、高效的生产提供了技术支持。 |

Abstract: |
【Objective】Process conditions for Lipase catalyzed synthesis of conjugated linoleic acid phytosterol ester were optimized.【Method】Conjugated linoleic acid (CLA) phytosterol ester was synthesized by lipase Pseudomonas in water solution.Technological conditions were investigated and product was analyzed by gas chromatography (GC).On the basis of single factor experiments,reaction time,addition of enzyme,and reaction temperature were selected as the main factors influencing the esterification reaction.The reaction conditions were optimized by response surface methodology (RSM) using the degree of esterification (DE) as the evaluation index and quadratic regression equation was established and verified.【Result】Optimal conditions of synthesis of conjugated linoleic acid phytosterol ester were obtained:Reaction temperature 55.5 ℃,enzyme amount 6.6%,reaction time 41.2 h,and DE 84.37%.【Conclusion】Investigation on Lipase catalyzed synthesis of conjugated linoleic acid phytosterol ester provided technical support for safe and efficient production of phytosterol ester. |
