僵蚕乙醇提取物对HearNPV的增效作用研究

摘要:
【目的】利用僵蚕乙醇提取物作为棉铃虫核型多角体病毒(HearNPV)的生物增效剂,研究其对棉铃虫核型多角体病毒(HearNPV)毒力的增效作用与内在机制。【方法】以棉铃虫3龄幼虫为标靶试虫,在HearNPV中添加0~80 μg/mL僵蚕乙醇提取物,研究其对HearNPV的增效作用,并通过僵蚕乙醇提取物对试虫ALP活性的影响探讨其增效作用机理。【结果】僵蚕乙醇提取物对HearNPV具有显著的增效作用,能够显著提高病毒的毒力,40 μg/mL的剂量水平就能够使病毒感染的3龄棉铃虫在第6天达到90%的死亡率、第7天达到100%的死亡率。剂量80 μg/mL的僵蚕乙醇提取物能够显著缩短棉铃虫3龄试虫的LT50和LT90值,其LT50和LT90分别较对照缩短23.2%和27.8%。同时,在HearNPV中添加僵蚕乙醇提取物可以显著降低试虫中肠的ALP活性,僵蚕乙醇提取物的使用剂量达到40 μg/mL以上时,试虫中肠ALP活性较对照显著下降,且差异随感染时间的延长进一步增大。【结论】僵蚕可以作为病毒增效剂新材料,其在克服传统杆状病毒制剂杀虫速度慢的缺陷方面具有极好的发展前景。

Abstract:
【Objective】Ethanol extract of Bombyx batryticatus (BB extract) was used as biological virulence enhancer for Helicoverpa armigera nucleopolyhedrovirus (HearNPV).【Method】BB extract was used as virulence enhancer for HearNPV against 3 instar larvae of H.armigera.The dose of BB extract was 0-80 μg/mL.The possible mechanism of virulence enhancement by BB extract was investigated by ALP enzyme assay.【Result】The mortalities at 6 days and 7 days after infection with 40 μg/mL BB extract reached 90% and 100%,respectively.80 μg/mL BB extract enhancer significantly reduced LT50 and LT90 values by 23.2% and 27.8%,respectively compared to the pure HearNPV infection.Meanwhile,ALP activity of the tested larvae was inhibited dramatically when the concentrations of BB extract were >40 μg/mL,and the inhibition increased gradually.【Conclusion】BB extract was a potentially novel virulence enhancer with a great prospect.It could overcome the shortages of baculoviruses based traditional bio pesticides.
