中药复方对免疫抑制模型小鼠细胞因子、免疫球蛋白及红细胞免疫黏附功能的影响

摘要:
【目的】评价自拟中药复方对免疫抑制小鼠血清细胞因子、免疫球蛋白和红细胞免疫黏附功能的影响。【方法】通过腹腔注射环磷酰胺建立免疫抑制小鼠模型。试验组免疫抑制小鼠分别按20,10和5 g/kg剂量灌胃自拟中药复方,同时设置模型对照组(灌胃等体积生理盐水)、健康对照组(灌胃等体积生理盐水)和黄芪多糖对照组(灌胃剂量66 mg/kg),连续7 d,试验第0,3,7天采集血样,用血细胞自动分析仪测定小鼠血细胞数,用酶联免疫吸附试验测定血清IL-2、IgG、IgM、IL-4和IFN-γ含量,并计算IL-4/IFN-γ,用红细胞C3b受体花环(C3bRR)及免疫复合物花环(ICR)试验分析红细胞免疫黏附功能。【结果】通过腹腔注射环磷酰胺(80 mg/kg,1次/d,连用3 d)成功建立了免疫抑制小鼠模型。所用中药复方和黄芪多糖可显著提高免疫抑制模型小鼠WBC,其中以10和20 g/kg剂量中药复方效果显著;10和20 g/kg剂量中药复方可显著提高免疫抑制模型小鼠血清中IgG和IgM含量,提高或恢复免疫抑制模型小鼠血清中IL-2含量,并可促进免疫抑制模型小鼠血清IL-4和IFN-γ含量、IL-4/IFN-γ、红细胞C3bRR率恢复至健康小鼠水平;所用中药复方和黄芪多糖对免疫抑制模型小鼠红细胞ICR率的影响不明显。【结论】自拟中药复方通过调控细胞因子和免疫球蛋白生成,对环磷酰胺介导免疫抑制模型小鼠的细胞免疫、体液免疫和红细胞免疫黏附功能发挥正向调节作用,其中以10 g/kg剂量效果明显。

Effects of Chinese herbal compound on cytokines,immunoglobulins and erythrocyte immune adhesion function of immunosuppressive mice

Abstract:
【Objective】The present study aimed to evaluate effects of self-made Chinese herbal compound on cytokines,immunoglobulins and erythrocyte immune adhesion function of immunosuppressive mice. 【Method】Immunosuppressive model mice established by intraperitoneal injection of cyclophosphamide (CY),were treated with different doses of Chinese herbal compound (20,10 and 5 g/kg per day) as treatments,Astragalus polysaccharide (66 mg/(kg·d)) as drug positive control,and distilled water (0.2 mL/mouse every day) as model control while healthy mice were treated with distilled water (0.2 mL/mouse every day) as the negative control.The experiment last 7 days and blood samples were taken before the treatment,and the third day and the seventh day after the treatment to count white cell numbers with automatic blood cell analyzer,detect contents of interleukin-2 (IL-2),interleukin-4 (IL-4) and interferon-γ (INF-γ) in serum with PR 521 analyzer through ELISA method,and evaluate erythrocyte adhesion function by erythrocyte C3b receptors (C3b-RR) and immune complex rosette test (ICR).【Result】Immunosuppressive model mice were successfully induced by injection of 80 mg/kg CY for three days and once per day.Both Chinese herbal compound and Astragalus polysaccharide significantly increased WBC counts in blood of immunosuppressive mice,and two groups treated by 20 and 10 g/kg doses of the Chinese herbal compound had the best results.Chinese herbal compound at the doses of 10 and 20 g/kg significantly affected contents of immunoglobins IgG and IgM,cytokines IL-2,IL-4,and IFN-γ,and ratios of IL-4 to INF-γ in serum of immunosuppressive mice.The rates of red cell C3b-RR were recovered to the healthy levels as well.In addition,Chinese herbal compound and Astragalus polysaccharide had no significant effects on ICR rates of red cells in the immunosuppressive mice.【Conclusion】The present Chinese herbal compound had positive effects on retrieve of cell immunity,humoral immunity and erythrocyte adhesion function of the immunosuppressive mice induced by CY via regulation of cytokines and immunoglobulins.The dose of 10 g/kg was suggested.
