DEHP对中国林蛙肝细胞中雌激素受体和卵黄蛋白原合成的影响

摘要:
【目的】探讨邻苯二甲酸二(2-乙基)己酯(di-2-ethylhexyl phthalate,DEHP)对诱导雄性两栖动物肝细胞合成卵黄蛋白原(Vitellogenin,VTG)的影响。【方法】将中国林蛙(Rana chensinensis) 雄性成体分别暴露在含10-7,10-6,10-5,10-4 mol/L DEHP的水体中,以自来水为对照,并分别于暴露20,30和40 d时取肝脏制作石蜡切片,用免疫组织化学方法检测肝细胞中雌激素受体(Estrogen receptor,ER)和VTG的表达情况。【结果】用不同浓度DEHP处理后,林蛙肝细胞中ER和VTG的相对表达量均显著或极显著高于对照组,且其相对表达量呈现随DEHP浓度的增高而增大的趋势,但二者的相对表达量随暴露时间延长而产生变化的规律性不明显。【结论】ER和VTG表达水平的一致性,揭示DEHP可通过诱导雄性两栖动物肝细胞中ER表达的增高而促使VTG合成。

Effects of di-2-ethylhexyl phthalate exposure on estrogen receptor and vitellogenin synthesis in hepatocytes of male frogs Rana chensinensis

Abstract:
【Objective】This study aimed to investigate the effects of di-2-ethylhexyl phthalate (DEHP) on the vitellogenin (VTG) synthesis in hepatocytes of male amphibians.【Method】Adult male frogs Rana chensinensis were exposed to 10-7,10-6,10-5 and 10-4 mol/L DEHP consecutively and their livers were taken at 20,30 and 40 d,respectively.The expressions of estrogen receptor (ER) and VTG protein in hepatocytes were detected by immunohistochemistry.【Result】 The expression levels of ER and VTG protein increased significantly in the hepatocytes of R.chensinensis exposed to DEHP compared to the control (Water).The relative expression levels of ER and VTG increased as the increase of DEHP dose at the same exposure time.When the concentration of DEHP was identical,VTG synthesis and expression of ER did not significantly fluctuate with extension of exposure.【Conclusion】The expression levels of ER with VTG protein were consistent,suggesting that DEHP could promote VTG synthesis by inducing the overexpression of ER in hepatocytes of male R.chensinensis.
