鼠隐孢子虫感染犊牛模型的建立及其血液生化指标的变化

摘要:
【目的】研究犊牛感染鼠隐孢子虫后血液生化指标、营养物质代谢相关酶及激素的变化,初步探讨隐孢子虫病的致病机理。【方法】给犊牛腹腔注射地塞米松磷酸钠注射液致其免疫力低下,然后通过经口感染鼠隐孢子虫卵囊构建隐孢子虫感染犊牛动物模型。采集模型组和对照组犊牛隐孢子虫感染前和感染后第14、28天的血液,测定血液中K+、Na+、Cl、Ca2+、Mg2+、血糖的浓度,以及总蛋白、白蛋白、5 羟色胺、白介素 2的质量浓度和酸性磷酸酶、碱性磷酸酶、谷草转氨酶、谷丙转氨酶的活性。【结果】经口感染鼠隐孢子虫卵囊犊牛粪便中检出了大小、形态与感染卵囊基本相同的隐孢子虫卵囊,表明隐孢子虫感染犊牛动物模型构建成功。隐孢子虫感染前,2组犊牛所有血液生化指标均无显著差异。与对照组相比,在犊牛感染隐孢子虫后14 d,血糖浓度、5-羟色胺质量浓度及碱性磷酸酶、谷草转氨酶活性均差异显著(P<0.05);感染隐孢子虫后28 d,血糖浓度,白介素-2、5-羟色胺质量浓度及碱性磷酸酶、谷草转氨酶和谷丙转氨酶活性均发生显著性变化(P<0.05)。【结论】成功构建了鼠隐孢子虫感染犊牛动物模型。隐孢子虫感染对犊牛营养物质的吸收利用有一定影响;与营养物质代谢相关的酶与激素对犊牛感染隐孢子虫后营养物质的吸收利用具有一定的调控作用。

Model establishment of calf infected with Cryptosporidium muris and the diversification of biochemical indexes in calf blood

Abstract:
【Objective】This study investigated the changes in biochemical indexes,and nutrition metabolism related enzymes and hormones in blood of calves infected with Cryptosporidium muris.【Method】Firstly,dexamethasone sodium phosphate injection was injected to calves to reduce immunity,then C.muris oocyst was orally infected to establish calf model with infection of C.muris.Blood samples were collected from calves in model group and control group before infection and 14 d and 28 d after infection.Concentrations of K+,Na+,Cl,Ca2+,Mg2+,blood sugar,total protein,albumin,IL-2 and 5-HT,and activities of acid phosphatase,alkaline phosphatase,aspartate aminotransferase and alanine aminotransferase were determined.【Result】C.muris oocyst with the same size,shape as oocyst used to infect calves was detected in the stool,indicating that the calf model with infection of C.muris was successfully established.Before the infection,blood biochemical indexes of calves in the two groups were not significantly different before infection.Compared with the control group,the contents of blood sugar and 5-HT and the activities of alkaline phosphatase and aspartate aminotransferase showed significant difference 14 d after infection (P<0.05).The contents of IL-2 and 5-HT and the activities of alkaline phosphatase,aspartate aminotransferase,and alanine aminotransferase showed significant difference 28 d after infection (P<0.05).【Conclusion】The calf model with infection of C.muris was successfully established.Infection of C.muris affected the nutrient absorption and utilization of calves and nutrition metabolism enzymes and hormones had certain regulation effects on the nutrition absorption and utilization after the infection of C.muris.
