WWP1与Myocardin结合调节血管平滑肌细胞的分化研究

摘要:
【目的】探讨含有WW结构域的E3泛素蛋白连接酶1(WWP1)在平滑肌细胞分化过程中的表达情况及其作用机制。【方法】用实时定量PCR,检测WWP1在人、小鼠主动脉血管平滑肌细胞中以及小鼠胚胎干细胞向平滑肌细胞体外分化过程中表达量的变化情况。用萤光素酶活性试验及免疫共沉淀试验检测WWP1与Myocardin的结合情况。【结果】WWP1在人和小鼠的血管平滑肌细胞中高表达,在全反式维甲酸诱导小鼠胚胎干细胞向平滑肌细胞分化过程中的表达水平明显上调。WWP1能与Myocardin蛋白结合,WWP1的表达水平受Myocardin调控。【结论】WWP1很可能通过与Myocardin结合参与血管平滑肌细胞分化的调节。

Regulation of WWP1 and Myocardin on differentiation of smooth muscle cell
Abstract:
【Objective】This study discovered the expression of WWP1 in the differentiation of smooth muscle cells and investigated the related mechanism.【Method】The mRNA expression level of WWP1 in human and mouse smooth muscle cells and the change of WWP1 expression level in smooth muscle cell differentiation were tested by real-time PCR.The binding interaction of WWP1 and Myocardin was tested by luciferase assay and confirmed by co-immunoprecipitation.【Result】WWP1 was highly expressed in both human and mouse smooth muscle cells.The expression level of WWP1 was up-regulated in the differentiation process of retinoid acid (RA) induced smooth muscle cell from mouse embryonic stem cells.WWP1 could bind and interact with Myocardin in protein level.The expression level of WWP1 was regulated by Myocardin.【Conclusion】WWP1 may play a role in smooth muscle cell differentiation through the binding interaction with Myocardin.
