双酚 A 对泥鳅肝组织结构及谷丙转氨酶活性的影响

摘要:
【目的】探讨双酚A(bisphenol A,BPA)对泥鳅(Misgurnus anguillicaudatus)肝组织结构和谷丙转氨酶(glutamic-pyruvic transaminase,GPT)活性的影响,为评价酚类化合物对水生生物的毒害作用提供理论依据。【方法】根据预试验确定的BPA对泥鳅的安全质量浓度和最小不致死质量浓度,按等对数(常用对数)法将BPA质量浓度设置4个(1.9,2.3,2.7,3.0 mg/L)处理,以未用BPA处理组为对照,分别在染毒7,14,21,28 d 取样,采用 H.E 染色法观察泥鳅肝组织结构的变化,采用赖氏比色法测定泥鳅肝脏GPT活性的变化。【结果】在1.9 mg/L BPA中暴露28 d时,泥鳅肝细胞肿大、空泡化及细胞溶解;在3.0 mg/L BPA 中仅暴露14 d时,泥鳅肝细胞除严重肿大、空泡化及细胞溶解外,还出现核变形与核溶解等受损症状。与对照组相比,4个BPA质量浓度组泥鳅在暴露各个时期GPT活性均有明显升高,但随着BPA质量浓度的增大,GPT活性逐渐降低;在同一BPA质量浓度下,随着暴露时间的延长,GPT活性呈现先升高后降低的趋势。【结论】BPA可造成泥鳅肝组织结构损伤,并随BPA质量浓度的增大而加剧;BPA对泥鳅肝脏GPT的活性具有显著影响,可导致肝功能受损

Effects of bisphenol A on hepatic tissue structure and GPT activities in Misgurnus anguillicaudatus
Abstract:
【Objective】This paper studied the effects of bisphenol A (BPA) on hepatic histopathology and GPT activities of Misgurnus anguillicaudatu to improve the evaluation of phenolic compounds toxicity to hydrobionts.【Method】Base on the safe concentration and the minimal non-lethal concentration of BPA to M.anguillicaudatus in preliminary tests,four cases with BPA mass concentrations of 1.9,2.3,2.7 and 3.0 mg/L were set up,respectively.Hepatic tissues at 7,14,21 and 28 d in M.anguillicaudatus were observed using H.E method and GPT activities were determined using Lai’s colorimetric method.【Result】The hepatic cells exposed to 1.9 mg/L BPA for 28 d were enlarged and bubbled,and cell lysis was observed,while karyolysis and nucleus deformation were observed in addition to swelling,vacuolization and lysis on that exposed to 3.0 mg/L BPA for 14 d.GPT activities of cells exposed to different BPA mass concentrations increased significantly compared to control.However,the GPT activities decreased as the increase of exposure BPA concentrations,and the GPT activities increased initially followed by decrease as the increase of exposing time at same BPA mass concentrations.【Conclusion】BPA damaged hepatic structure and the damages aggravated as the increase of exposure concentrations.BPA significantly affected hepatic GPT activities of M.anguillicaudatus,and damaged liver function.
