不同鸭种胚胎期和岀雏早期肌肉组织中IGF-Ⅰ基因mRNA的差异表达分析

摘要:
【目的】研究不同品种鸭胚胎期和出雏早期胸肌、腿肌中IGF-Ⅰ基因mRNA表达水平的发育性变化。【方法】采用实时荧光定量PCR方法,研究生长速度不同的“高邮鸭”和“金定鸭”13,17,21,25,27胚龄和出雏后7日龄胸肌、腿肌中IGF-Ⅰ基因mRNA的表达规律。【结果】IGF-Ⅰ mRNA的表达规律在2个品种鸭的早期肌肉发育过程中较为一致,胸肌和腿肌IGF-ⅠmRNA表达量都是在13胚龄时最高,公鸭和母鸭同一肌肉组织IGF-Ⅰ mRNA表达没有显著差异(P>0.05);不同品种间,IGF-Ⅰ mRNA在“高邮鸭”胸肌中的表达量除27胚龄外,其他发育时期均高于“金定鸭”,且在13和21胚龄及7日龄时呈显著(P<0.05)或极显著差异(P<0.01),而“高邮鸭”腿肌IGF-Ⅰ mRNA的表达量在不同发育时期均高于“金定鸭”,但差异不显著(P>0.05);同一品种内IGF-Ⅰ mRNA在胸肌中的表达量高于腿肌,且除13胚龄外,其他发育时期都显著(P<0.05)或极显著(P<0.01)高于其在腿肌中的表达量。【结论】鸭肌肉中IGF-Ⅰ基因mRNA表达水平具有显著的年龄依赖性和明显的品种特征及组织差异,肌肉中IGF-Ⅰ 基因mRNA表达对鸭早期生长发育有重要影响,且对胸肌生长发育的启动要早于腿肌。

Expression analysis of IGF-Ⅰ gene mRNA in muscles of
Abstract:
【Objective】This experiment was conducted to study the change patterns of IGF-Ⅰgene mRNA expression in muscles of different ducks during embryonic and neonatal stages.【Method】Real time PCR was applied to compare the expression of insulin-like growth factor-I (IGF-Ⅰ) mRNA in pectorales and legs of meat-type duck breed-Gaoyou ducks and egg-type duck breed-Jinding ducks at the embryonic days of 13,17,21,25,and 27 and the 7th day after birth.【Result】The profiles of IGF-ⅠmRNA in muscles of two duck breeds were relatively consistent during early development.The highest level of IGF-Ⅰ mRNA appeared at embryo age of 13 day in pectorales and legs of both breeds,and there was no significantly difference (P>0.05) in same muscle tissues between male and female ducks.The expression level of IGF-Ⅰ mRNA in pectorals of Gaoyou ducks was higher than that in Jinding ducks except at embryo age of 27 day,and the differences at embryo age of 13 and 21 day and age of 7 day were significant (P<0.05 or P<0.01).The expression level of IGF-Ⅰ mRNA in legs of Gaoyou ducks was higher than that in Jinding ducks at different development stages (P>0.05).Within the same breed,the expression levels of IGF-Ⅰ mRNA in pectorals were significantly higher than that in legs except at embryo age of 13 day (P<0.05 or P<0.01).【Conclusion】The expression of IGF-Ⅰ gene mRNA was affected by age,breed and tissues.The expression of IGF-Ⅰ gene mRNA in muscles was important to the early growth and development of ducks and the expression in pectorals started earlier than in legs.
