短枝六道木叶果胶提取工艺优化及理化性质研究

摘要:
【目的】探明短枝六道木叶果胶的提取工艺及理化性质。【方法】在单因素试验的基础上,选取了提取温度、提取时间、pH值、液(mL)料(g)比4个因素进行Box-Behnken中心组合设计,利用响应面分析法对超声波辅助提取短枝六道木叶果胶工艺进行优化,并将最佳工艺条件下所得果胶与市售柑橘果胶的理化性质进行比较。【结果】提取短枝六道木叶果胶的最佳工艺参数为:提取温度75 ℃,pH值1.7,提取时间30 min,液料比1∶30,在此条件下果胶的理论得率为18.56%,实测得率为18.79%,相对误差为0.012 4。所得短枝六道木叶果胶酯化度为62.50%,半乳糖醛酸含量68.78%,溶解度和持油力分别为98.45%和1.98 g/g,DPPH清除率为78.81%,Fe3+还原力(FRAP)为5.13 mmol/L。【结论】最佳工艺条件下所提取的短枝六道木叶果胶属高甲氧基果胶,具有较强的持油力、溶解性和一定的抗氧化能力。

Optimization of extraction technology of Abelia engleriana leaves pectin and its physicochemical properties
Abstract:
【Objective】The objective of this study was to obtain the optimal conditions of ultrasound-assisted extraction and the physicochemical properties of the pectin of Abelia engleriana leaves.【Method】The effects of temperature,time,pH and liquid/solid on the yield of pectin were investigated with a four-factor Box-Behnken center-united design based on single-factor experiments.Then the comparisons of physicochemical properties between the extracts under the best optimal conditions and commercial citrus pectin (CP) were performed.【Result】The results showed that the optimal conditions of temperature,pH,time,and liquid/solid ratio were 75 ℃,1.7,30 min,and 1∶30,respectively.The predicted yield was 18.56% and observed yield was 18.79% with a relative error of 0.012 4.The pectin consisted of 68.78% galacturonic acid content with DE value of 62.50%,and had the solubility and oil holding capacity of 98.45% and 1.98 g/g,respectively.The DPPH free radical-scavenging activity and FRAP value were 78.81% and 5.13 mmol/L,respectively.【Conclusion】The pectin under the optimal conditions belonged to the high methoxyl pectin,and had a high solubility,high oil holding capacity,and certain antioxidant ability.
