党参粗多糖的组成及其免疫活性研究

摘要:
【目的】研究党参粗多糖的组成及其对小鼠主要免疫细胞活性的影响,为党参的开发利用提供依据。【方法】以东党参为试材,提取分离其粗多糖,利用气相色谱法对其单糖组成进行分析。制备小鼠(ICR雄性)脾脏B淋巴细胞悬液和纯化的巨噬细胞,分别应用四甲基偶氮唑盐微量酶反应比色法(MTT法)和中性红吞噬试验测定党参粗多糖对小鼠脾脏B淋巴细胞增殖和巨噬细胞吞噬功能的影响。【结果】党参粗多糖收率为9.7%,单糖组成为葡萄糖(Glc)、半乳糖(Gal)、木糖(Xyl)、鼠李糖(Rha)、阿拉伯糖(Ara)、半乳糖醛酸(GalA),其物质的量比为102.7∶7.9∶2.2∶1.8∶1.1∶1.0。党参粗多糖能够刺激小鼠脾脏B淋巴细胞增殖,并呈现一定的量效关系,当粗多糖质量浓度为400 μg/mL 时,可以显著促进小鼠B淋巴细胞的增殖(P<0.05);党参粗多糖可极显著增强小鼠巨噬细胞吞噬中性红的能力。【结论】本研究提取党参粗多糖的方法简单,提取的粗多糖可促进小鼠B淋巴细胞的增殖,提高巨噬细胞的吞噬功能,从而增强免疫细胞活性,有望成为新一代免疫调节剂。

Study on composition and immunological activities of crude polysaccharide isolated from the Codonopsis pilosula
Abstract:
【Objective】The research was to study compositional analysis and immunological activities of water soluble crude polysaccharide from the Codonopsis pilosula to offer theoretical basis for developing medicament.【Method】A water-soluble crude polysaccharide has been isolated from the root of Codonopsis pilosula,and compositional analysis of the crude polysaccharide was detected by GC.Lymphocytes from spleen cells and phagocytic cells were purified from male ICR mouse to study the influence of crude polysaccharide of Codonopsis pilosula on mouse immune cells by using MTT(3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide)assay and neutral red approach respectively.【Result】The crude polysaccharide was 9.7% isolated from the hot-water extract of Codonopsis pilosula.The results of GC analysis revealed that the monosaccharide of crude polysaccharide of Codonopsis pilosula was made up of Glc,Gal,Xyl,Rha,Ara and GalA with the molarity ratio of 102.7∶7.9∶2.2∶1.8∶1.1∶1.0.The results showed that crude polysaccharide of Codonopsis pilosula remained higher immunological competence on splenic lymphocytes even if the polysaccharide density achieved 400 μg/mL.Different density crude polysaccharide of Codonopsis pilosula could enhance the function of peritoneal macrophage engulfing neutral red and its metabolism.【Conclusion】The extraction process of crude polysaccharide of Codonopsis pilosula is simple,and the polysaccharide has marked immunity activities and will be a good candidate to develop immunity conditioner.
