长角豆组织培养繁育技术研究

摘要:
【目的】探讨长角豆“卡苏达”(Ceratonia siliqua L.‘Casuda’)的组织培养快速繁育技术,为其在我国的成功引种及种质资源保存奠定基础。【方法】以引自西班牙的长角豆“卡苏达”种子为外植体,培养无菌苗后,取其茎段作为组培材料,在筛选出初代基本培养基的基础上,又进行了继代增殖培养基和生根诱导培养基的筛选,最后对长角豆组培生根苗进行了炼苗和移植。【结果】长角豆种子无菌苗在DKW+1.0 mg/L 6-BA+0.1 mg/L IBA+25 g/L蔗糖的增殖培养基上生长最好;最适继代增殖培养基为DKW+0.8 mg/L 6-BA+0.1 mg/L IBA+60 mg/L L 赖氨酸+25 g/L蔗糖;最佳生根培养基为1/2 DKW+0.4 mg/L IBA+20 g/L蔗糖,生根率为93.33%,生根苗移栽后的平均成活率达到92.36%。【结论】成功建立了长角豆的组织培养再生体系,为长角豆“卡苏达”的快速繁育提供了技术支持。

Abstract:
【Objective】The study was to explore the tissue culture and rapid propagation technology of the Ceratonia siliqua L.‘Casuda’,which would lay a basis for the successful introduction and storage of germplasm resources.【Method】The C.siliqua L.‘Casuda’ seeds from Spain were taken as explants,then the aseptic seedlings were cultured.The stem segments of the aseptic seedlings were taken as materials,after the primary basic mediums screening,screening the secondary proliferation culturing mediums and roots induction mediums was performed,finally,the aseptic seedlings were practiced and transplanted.【Result】In the primary culture period,the most appropriate medium was DKW+1.0 mg/L 6-BA+0.1 mg/L IBA+25 g/L sugar.DKW+0.8 mg/L 6-BA+0.1 mg/L IBA+60 mg/L L-lysine+25 g/L sugar for subculture.The recommended medium for rooting stage was 1/2 DKW+0.4 mg/L IBA+20 g/L sugar,and the rooting percentages were up to 93.33%.The average survival rate of transplanting was up to 92.36%.【Conclusion】The tissue culture regeneration system was established,which provides technical support for rapid multiplication of Ceratonia siliqua L.‘Casuda’.
