雌激素和IFN-γ对去卵巢大鼠下丘脑中Bcl-2和Bax表达的影响

摘要:
【目的】研究雌激素与γ-干扰素(IFN-γ)对下丘脑的协同保护作用。【方法】建立去卵巢(OVX)模型大鼠,然后将其分为摘除卵巢(OVX)组、假手术(SHAM)组、OVX+17-β雌二醇(E2)组、OVX+E2+50 IU IFN-γ组、OVX+E2+200 IU IFN-γ组和OVX+E2+800 IU IFN-γ组共6组,分别在处理后的0,2,4,6,8周,用免疫组织化学SP法对各组大鼠下丘脑中凋亡相关蛋白Bcl-2和Bax的表达进行检测。【结果】OVX组大鼠下丘脑中Bcl-2的相对表达量随时程逐渐减少,4周后最低,6周后又逐渐上升,随后维持在一定的水平,Bax表达量的变化趋势则与之相反;OVX组Bcl-2和Bax的表达量与SHAM组差异极显著(P<0.01)。OVX+E2组大鼠下丘脑中Bcl-2的相对表达量增加,Bax的相对表达量降低,均与OVX组差异显著(P<0.05)。OVX+E2+200 IU IFN-γ组Bcl-2的相对表达量极显著高于OVX组,显著高于OVX+E2组,Bax的总体变化趋势与OVX+E2组一致;OVX+E2+50 IU IFN-γ组和OVX+E2+800 IU IFN-γ组Bcl-2和Bax的相对表达量与OVX+E2组差异不显著(P>0.05)。【结论】雌激素的缺乏增强了下丘脑神经元对凋亡的敏感性,而外源生理剂量E2与适当剂量的IFN-γ,在一定时期内对这种凋亡有重要的保护作用。

Effect of estrogen and IFN-γ on the expression of Bcl-2 and Bax in hypothalamus of ovariectomed rats
Abstract:
【Objective】The research was to study the synergy effect of estrogen and interferon-γ(INF-γ) on protecting the hypothalamus nucleus.【Method】The immunohistochemical SP method was used to study the expressions of Bcl-2 and Bax in hypothalamus of the ovariectomized (OVX) group,the SHAM group,the OVX+17-β estrogen(E2) group,OVX+E2+50 IU IFN-γ group,OVX+E2+200 IU IFN-γ group and OVX+E2+800 IU IFN-γ group.【Result】The main results were as follows:After ovariectomization,relative expressions of Bcl-2 positive productions(μ2) tapered with the course and reached the minimum after 4 weeks concurrently.The μ2 of Bax increased and reached the maximum after 4 weeks,which had significant difference with the SHAM group(P<0.01).OVX+E2 group,the μ2 of Bcl-2 increased and had remarkable difference with OVX group(P<0.01),the μ2 of Bax decreased and had remarkable difference with OVX group(P<0.01);The μ2 of Bcl-2 of OVX+E2+200 IU IFN-γ group had different changes with OVX group(P<0.01),OVX+E2 group(P<0.05),and SHAM group(P>0.05).The total tendency of the μ2 of Bax had very significant difference with OVX group(P<0.01) in the nucleus which the μ2 of Bax decreased.【Conclusion】The sensibility of the neurons in hypothalamus to apoptosis was enhanced by estrogen deficiency,and physiological dose E2 combined with proper-dose IFN-γ had important protecting effect on the apoptosis within a certain period.
