埃里格孢菌FEL3产SW发酵培养条件的优化

摘要:
【目的】对埃里格孢菌(Undifilim oxytropis)FEL3产苦马豆素(SW)的发酵条件进行优化。【方法】在不同碳源(葡萄糖、蔗糖、乳糖、燕麦片、可溶性淀粉)、氮源(豆粉、酵母膏、牛肉膏、蛋白胨、硝酸钠、氯化铵、硫酸铵)、无机盐(MgSO4·7H2O,FeSO4·H2O,ZnSO4·H2O,CaCl2·2H2O,CuSO4·5H2O,KH2PO4)、初始pH(4.5,5.0,5.5,6.0,6.5,7.0,7.5)和接种量(体积分数2%,4%,6%,8%,10%)培养条件下,将埃里格孢菌FEL3接种到液体培养基中,培养15 d后,测定菌丝湿、干质量浓度和菌液中SW浓度,选择最佳培养条件。【结果】当碳源为燕麦片、氮源为豆粉、无机盐为CuSO4·5H2O、培养基初始pH为6.5~7.0、接种量为体积分数8%时,埃里格孢菌FEL3菌丝湿、干质量浓度和菌液中SW浓度均较高。【结论】得到了埃里格孢菌FEL3产SW的最佳培养条件,为用埃里格孢菌 FEL3大量生产SW奠定了基础。

Optimization of fermentation conditions for producing SW by Undifilim oxytropis FEL3
Abstract:
【Objective】The culture conditions of Undifilim oxytropis FEL3 for producing SW were optimized.【Method】The U.oxytropis FEL3 were cultured under the different carbon sources (glucose,sucrose,lactose,oatmeal and tragantine),nitrogen sources (soya flour,yeast extract,beef extract,proteose peptone,sodium nitrate,ammonium chloride and ammonium sulfate),inorganic salts (MgSO4·7H2O,FeSO4·H2O,ZnSO4·H2O,CaCl2·2H2O,CuSO4·5H2O,KH2PO4),initial pH(4.5,5.0,5.5,6.0,6.5,7.0,7.5) and inoculation volumes (2%,4%,6%,8%,10%) for 15 days.The culture conditions were optimized by detection of wet weight concentration,dry weigh concentration of mycelia and SW concentration in culture medium.【Result】The wet weight concentration,dry weigh concentration of mycelia and SW concentration in culture medium were higher when oatmeal was used as carbon source,soybean meal as nitrogen source,CuSO4·5H2O as inorganic salt,and the optimum initial pH was 6.5-7.0,quantity of microorganism inoculated volume fraction 8%.【Conclusion】The study obtained the best culture conditions of U.oxytropis FEL3 for producing SW,and lay a basis for mass-production of SW by the U.oxytropis FEL3.
