【目的】研究牛肌细胞生成素基因(MyoG)第2、3外显子及其侧翼区单核苷酸的多态性(SNPs),分析其与牛部分体尺性状的相关性。【方法】以6个牛群体(鲁西牛、鲁杂牛(鲁西牛×西门塔尔牛)、南阳牛、夏南牛、郏县红牛、秦川牛)共779头3~4岁左右母牛为研究材料,通过PCR-SSCP和DNA测序技术检测牛MyoG基因上存在的SNPs,并分析其与牛部分体尺性状的关联性。【结果】MyoG基因第2外显子及其侧翼区的扩增片段中不存在多态性,第3外显子及其侧翼区的扩增片段中存在3种基因型(AA、AB、BB),测序结果表明该突变位于3′-UTR 2 109位点处。最小二乘法分析表明,各群体中AA基因型个体的体斜长均显著高于BB基因型个体(P<0.05);鲁西牛群体中AA基因型个体的尻长显著高于BB基因型(P<0.05);郏县红牛群体中AA基因型个体的体高显著高于BB基因型(P<0.05)。3种基因型对其他体尺性状没有显著影响(P>0.05)。【结论】MyoG基因对黄牛体尺性状有一定影响,可作为黄牛体尺性状标记辅助选择的候选基因。 |

Single nucleotide polymorphism of MyoG gene in six cattle populations and its association with body measurement traits |

【Objective】This research was to study the polymorphism in exon 2 and 3 and their flanking regions of MyoG gene,and to analyze its relationship with several body measurement traits of cattle.【Method】779 female individuals aged 3-4 years from six populations (Luxi,Luxi×Simmental crossbred steers,Nanyang,Xia’nan,Jiaxian red and Qinchuan) were selected to detect polymorphism of MyoG gene by using PCR-SSCP and DNA sequencing technique,and to study its association with part body measurement traits.【Result】The results indicated that there was not polymorphism in exon 2 and its flanking region and there were three genotypes (AA,AB,BB) detected by exon 3 and its flanking region.The sequencing results showed that the mutation of A>G was detected in 3′-UTR of the bovine MyoG gene.Statistical analysis indicated that in all populations,animals with AA genotype have higher mean values for body length than those with the BB genotype (P<0.05);in Luxi population,animals with the AA genotype have higher mean values for rump length than those with the genotype BB (P<0.05);in Jiaxian red population,animals with the AA genotype have higher mean values for body height than those with the BB genotype (P<0.05).Genotype analysis did not show any significant effect on other traits(P>0.05).【Conclusion】It was concluded that MyoG gene has some effect on body measurement traits and can be used as a candidate gene for marker-assisted selection in cattle. |
