靛玉红-3′-单肟对猕猴成纤维细胞增殖和凋亡及周期同步化的影响

摘要:
【目的】研究靛玉红-3′-单肟(Indirubin-3′-monoxime)对猕猴成纤维细胞增殖、凋亡和周期同步化的影响。【方法】用PI和FITC同时对细胞进行染色,用流式细胞仪检测荧光强度,对细胞内DNA和蛋白质的含量作相对定量,分析细胞在G0,G0+G1,S,G2+M期的分布比例;用BrdU标记法检测靛玉红-3′-单肟处理对细胞增殖的影响,用原位末端标记法(TUNEL)检测细胞凋亡的发生情况。【结果】(1)与处于对数增殖期(对照)的细胞相比,10 μmol/L靛玉红-3′-单肟处理30 h后,G0与G0+G1期细胞比例分别由7.27%和84.26%增加为14.21%和94.22%,差异达显著水平。(2)与对数增殖期细胞(BrdU阳性率为80%)相比,靛玉红-3′-单肟处理30 h可显著地抑制细胞的增殖,在后续标记培养24 h期间只有8.15%的细胞呈BrdU阳性,如果是在去除药物的正常条件下标记培养24 h,大多数细胞(58.28%)又可进入增殖状态。(3)与正常培养对照组细胞凋亡率(1.12%)相比,靛玉红-3′-单肟处理组凋亡发生率为1.82%,虽有所增加但差异不显著。【结论】靛玉红-3′-单肟是一种可逆的细胞周期抑制剂,对猕猴成纤维细胞增殖的抑制作用是可逆的,对细胞凋亡发生率没有显著影响。

Effects of indirubin-3′-monoxime on cell proliferation,apoptosis and cycle synchronization in rhesus monkey fibroblasts
Abstract:
【Objective】This study was to investigate the effects of indirubin-3′-monoxime on proliferation,apoptosis and cycle synchronization of rhesus monkey fibroblasts.【Method】Cells were stained with PI and FITC,then were analyzed with flow cytometry to detect the distribution of cells in G0,G0+G1,S,and G2+M stages.Cell proliferation was analyzed with BrdU incorporation into new synthesized DNA synthesis.Apoptosis was detected with in situ TUNEL analysis.【Result】(1)Compared with cycling cells,proportions of G0 and G0+G1 cells significantly increased from 7.27% and 84.26% to 14.21% and 94.22% in cultures treated with 10 μmol/L indirubin-3-monoxime.(2)During 24 h BrdU labeling culture,cell proliferation was significantly inhibited and only 8.15% of indirubin-3′-monoxime treated cells were BrdU-positive in contrast to 80% of cycling control cultures.However,after removal of indirubin-3′-monoxime,58.28% cells reentered proliferation during 24 h labeling.(3)Apoptosis occurrence increased,but not significantly,in cultures treated with indirubin-3′-monoxime (1.82%) compared with normal control cultures (1.12%).【Conclusion】Indirubin-3′-monoxime is a kind of reversible cycle inhibitor for rhesus monkey fibroblasts,which can inhibit cell proliferation in a reversible way and does not induce apoptosis.
