蒺藜苜蓿小G蛋白MtROP5对拟南芥角果及侧器官发育的影响

摘要:
【目的】利用拟南芥异源表达MtROP基因并研究其功能, 为蒺藜苜蓿中MtROP蛋白的功能研究,及进一步探索MtROP蛋白在蒺藜苜蓿和微生物共生互作过程中的作用提供理论依据。【方法】利用同源克隆方法获得了蒺藜苜蓿(Medicago tructula)中与拟南芥AtROP5高度同源的ROP蛋白基因MtROP5,用MtROP5表达载体转化拟南芥并严格筛选转化植株,观察T-2代转化植株角果、侧枝分叉、簇毛等器官的表型,并结合启动子GUS、GFP、RT-PCR等试验手段,研究MtROP5的组织表达特性及其对各种激素的应答反应,通过拟南芥异源表达MtROP5对其功能进行研究。【结果】与对照相比,过表达CAMtROP5(Constitute activity MtROP5,CAMtROP5)、MtROP5、GFP -MtROP5的拟南芥植株的角果变短,而角果数目相应增加。过表达CA MtROP5植株角果的顶端具有较多花粉管,DN-MtROP5(Dominant negative MtROP5,DN-MtROP5)及MtROP5转化植株的花粉管则较早凋亡。过表达CA-MtROP5、DN-MtROP5植株的侧生器官及花序的极性发生改变,转化CA-MtROP5和MtROP5的转基因拟南芥的簇毛分支数目增加。启动子GUSGFP-MtROP5的表达模式表明,该基因在叶脉、花药、下胚轴、气孔及簇毛等部位特异表达。RT-PCR及启动子GUS表达分析表明,生长激素可诱导该基因的上调表达。【结论】MtROP5在果实及侧器官的极性发育中具有较为重要的作用,可能参与了生长素相关途径的调控。

Effects of over expression of a Medicago truncatula small G protein MtROP5 on the development of silique and lateral organ in Arabidopsis thaliana
Abstract:
【Objective】The objective was to study the function of ROP(Rho GTPase of plant) gene in Medicago truncatula by transforming various constructs into Arabidopsis thaliana to provide theoretical basis for further investigation of the function of ROP gene in M.truncatula and the function of MtROP gene in the process of symbiotic interaction between host legume plant and rhizobium.【Method】One small G protein which has high homology with AtROP5 was cloned by using the pathway of homologus cloning in M.truncatula,and was named MtROP5.The function of MtROP5was studied by transforming it into Arabidopsis.【Result】Results showed that over expression of CA-MtROP5(Constitute activity MtROP5,CA-MtROP5),MtROP5,GFP-MtROP5 can shorten the length of silique of Arabidopsis,and the stigma of plants transformed with CA-MtROP5 has more pollen tubes than that transformed with vector,DN-MtROP5 (Dominant negative MtROP5,DN-MtROP5) and MtROP5.The polarity of lateral organ and anthotaxy and the polarity of trichome change after transformation with various construnction of MtROP5.The expression pattern of the promoter of MtROP5 fused GUS and GFP-MtROP5 showed that MtROP5 is the specific expression in vein,anther,hypocotyl,stoma and trichome.The analysis of RT-PCR and the promoter of MtROP5 fused GUS after treated with various plant hormone showed that the expression of MtROP5 is up regulated by auxin.【Conclusion】MtROP5 plays a critical role in silique and lateral organ development,and may be involved in the regulation of auxin signal pathway.
