| 【目的】研究通爽冲剂对便秘的疗效并初步探索其作用机制。【方法】用复方地芬诺酯建立小鼠便秘模型，以正常对照组、便秘模型组、麻仁丸组、西沙必利组作为对照，观察低（6.44 g/kg）、中（12.87 g/kg）、高（19.31 g/kg）3个剂量的通爽冲剂，对模型小鼠排便时间、排便性状、墨汁推进率及含水率的影响，并取小鼠肠段制作石蜡切片进行HE染色，观察通爽冲剂对肠组织的影响。采用家兔空肠水囊植入的方法，以PCLAB-UE生物医学信号采集处理系统记录空肠运动曲线。向家兔的回肠和结肠注入试验药物，测量各肠段内的液量，并观察肠壁的肿胀程度和肠黏膜的充血程度。【结果】在成功制造便秘模型的情况下，通爽冲剂各剂量组小鼠排首粒黑便时间和粪便含水率与便秘模型组比较，组间两项数据均差异极显著（P＜0.01）；中、高剂量组小鼠排首粒黑便时间和粪便含水率与西沙必利、麻仁丸组比较，组间两项数据均差异极显著（P＜0.01），且粪便性状良好。在小肠和大肠的墨汁试验中，通爽冲剂各剂量组的墨汁推进率与便秘模型组比较差异极显著（P＜0.01）；中、高剂量组的墨汁推进率与麻仁丸、西沙必利组比较差异极显著（P＜0.01）。镜检HE染色切片显示，与正常组织比较，通爽冲剂3个剂量组均未对小鼠肠组织结构造成任何损害。灌服通爽冲剂后，家兔在体小肠运动幅度显著增加。家兔肠内药物灌注试验显示，通爽冲剂能使结肠及回肠肠内液量显著增加；经通爽冲剂作用的肠段与西沙必利组比较，无明显的充血、肿胀现象。【结论】通爽冲剂能有效增加肠动力，抑制肠管对水分的过度吸收，可使便秘模型排便形态、含水率正常，具有良好的通便作用，并且对肠道无刺激、无损害，对治疗便秘有很好的效果。 |

| 【Objective】The study was done to verify the therapeutic effect of TongShuang dissolved medicine on constipation and explore its mechanism of action.【Method】Compound diphenoxylate was used to create mice constipation model,and with normal group,constipation model group,Maren Pill group and cisapride group as controls,the effects of the 6.44 g/kg(low),12.87 g/kg (middle),19.31 g/kg(high) of TongShuang dissolved medicines on mice defecation time,defecation characters,ink forward rate and moisture content were observed,then the mouse intestines HE staining was taken to observe the effects of TongShuang dissolved medicines on intestinal tissue using Jejunum of rabbits implanted with Water bag,PCLAB-UE biomedical signal acquisition and processing system,and jejunum motion curve was recorded.The ileum and colon of rabbits were injected with the test drugs,the amount of fluids within intestines was measured and the degree of bowel wall swelling and mucosal congestion observed.【Result】In the constipation model,mice fed TongShuang dissolved medicine ranked the first,the amount of black stool was more than that of the model group and drug control groups and its traits were good,stool water content was better than that of cisapride group and Maren Pills group significantly;using microscopy to check HE stained glass slides,mouse used TongShuang dissolved medicine intestinal did not have any damage to the organizational structure compared with normal tissue;In the large intestine and small intestine,three dosage groups of TongShuang dissolved medicine were higher than that of the model group significantly (P<0.01),the ink promote rate of middle and high doses of TongShuang dissolved medicine group was higher than that of Maren Pills and cisapride group significantly (P<0.01).Compared with the normal group,all the concentrations of TongShuang dissolved medicine could enhance the contraction amplitude of in vivo small intestinal in rabbits respectively.After injection of TongShuang dissolved medicine,the amount of liquid inside intestines was more than that of the control group injected normal saline significantly.The intestinal mucosa injected Cisapride resulted in severe congestion and swelling,whereas TongShuang dissolved medicine groups appeared no significant changes.【Conclusion】TongShuang dissolved medicine can enhance the gastrointestinal motility effectively,inhibit intestinal absorption of excessive moisture,cure constipation model and form normal moisture content and cacation.TongShuang dissolved medicine has a good catharsis,and no irritable bowel,no damage to the bowel,so it has a good effect on constipation. |
