6个罗非鱼群体的遗传多样性及遗传关系研究

摘要:
【目的】探讨6个罗非鱼群体内的遗传多样性及群体间的遗传关系。【方法】利用20个微卫星标记分析6个罗非鱼群体的遗传多样性,研究其群体内遗传多样性和群体间遗传关系。【结果】19个微卫星位点扩增产物具有多态性,每个位点平均等位基因数为7.63;吉富尼罗罗非鱼、广东尼罗罗非鱼、美国尼罗罗非鱼和埃及尼罗罗非鱼遗传多样性较高,其平均杂合度分别为0.615 0,0.599 9,0.592 7和0.622 7,平均多态信息含量分别为0.607 0,0.530 2,0.509 5和0.520 5,平均有效等位基因数分别为2.866 3,3.132 1,2.649 5和3.366 8;而广东和美国2个奥利亚罗非鱼群体的遗传多样性较低,其平均杂合度分别为0.354 8,0.380 5,平均多态信息含量分别为0.281 7和0.280 7,平均有效等位基因数分别为1.933 8和1.707 7。UPGMA系统树分析结果表明,6个罗非鱼群体分为2支,其中2个奥利亚罗非鱼群体聚为一支,4个尼罗罗非鱼群体聚为一支。【结论】4个尼罗罗非鱼群体具有丰富的遗传多态性,而2个奥利亚罗非鱼群体的遗传多样性较低。19个微卫星标记均可用于罗非鱼群体的遗传多样性和系统发生关系的分析。

Analysis of genetic diversity in six tilapia populations
Abstract:
【Objective】The study was done to assess the genetic diversity and genetic relationship of nile tilapia Oreochromis niloticus and aureus tilapia Oreochromis aureus.【Method】The genetic diversity and genetic relationship of 278 tilapia from 6 populations were analyzed using 20 microsatellite markers.【Result】Polymorphic bands could be amplified in 19 pairs of primers and the average number of alleles in each pair of primers was 7.63.The genetic analysis of 6 tilapia populations showed that:Four nile tilapia populations of GIFT,Guangdong nile tilapia,American nile tilapia and Egyptian nile tilapia had a higher genetic polymorphism,the average heterozygosities were 0.615 0,0.599 9,0.592 7 and 0.622 7,respectively,and the average polymorphism information contents 0.607 0,0.530 2,0.509 5 and 0.520 5,respectively,and the average effective allele numbers 2.866 3,3.132 1,2.649 5 and 3.366 8,respectively.Two aureus tilapia populations of Guangdong and American had a low genetic polymorphism,the heterozygosities were 0.354 8,0.380 5,respectively,the average polymorphism information contents 0.281 7 and 0.280 7,respectively,and the average effective allele numbers were 1.933 8 and 1.707 7,respectively.NJ phylogenetic tree analysis showed that six populations were divided into two branches,the first branch consisted of the two aureus tilapia,and the second branch consisted of the four nile tilapia populations.【Conclusion】The four nile tilapia populations had a rich genetic polymorphism while the two aureus tilapia populations had a low genetic polymorphism.The 19 microsatellite markers were effective markers for the analysis of genetic diversity and relationships among tilapia populations.
