多杀菌素的提取和萃取条件研究

摘要:
【目的】 研究多杀菌素的最佳提取和萃取条件。【方法】 采用单因素试验,分别研究发酵液不同pH(6,7,8,9,10,11)、放置时间(2,4,8,16,24,32,48,72 h)和温度(20,30,40,50,60,70 ℃)对发酵液中多杀菌素提取效果及稳定性的影响,同时,从乙酸乙酯、石油醚、二氯甲烷和乙醚中筛选最适萃取试剂,并在此基础上研究萃取次数(1,2,3次)和萃取试剂体积对多杀菌素萃取效果的影响。【结果】 单因素试验结果显示,在发酵液pH为10时,多杀菌素提取效果较好;放置2~72 h,多杀菌素质量浓度不会发生显著变化;在20~70 ℃多杀菌素能稳定存在;乙酸乙酯为最适萃取试剂,最佳萃取次数为3次,其与多杀菌素提取液的最适体积比为1∶1。【结论】 得到了多杀菌素提取和萃取的最佳条件,在该条件下多杀菌素的质量浓度和萃取率分别为74.50 μg/mL和98.18 %。

Study on extraction conditions of spinosad in fermentation broth of Saccharopolyspora spinosa
Abstract:
【Objective】 The optimal extraction conditions of spinosad in fermentation broth of Saccharopolyspora spinosa were tested by parallel experiments.【Method】 Factors including pH(6,7,8,9,10,11), storage time(2,4,8,16,24,32,48,72 h),and temperature(20,30,40,50,60,70 ℃) on the effect of extraction results were investigated with one factor experiments.Meanwhile,the optimal extraction solvent was selected from ethyl acetate,petroleum ether,dichloromethane and ether.On the basis of the optimal extraction solvents,the extracting times (1,2,3) and extracting volume were optimized.【Result】 It was found that pH 10 was propitious to extract spinosad from fermentation broth.Spinosad in fermentation broth could be stored for 3 days at least within temperature from 20 ℃ to 70 ℃.Ethyl acetate was confirmed to the optimum extracting solvent,on the basis of which,three times was the optimal extracting times and the equivalent volume of ethyl acetate and fermentation extracting broth was the best extraction proportion.【Conclusion】 The optimal extraction conditions of spinosad in fermentation broth of Saccharopolyspora spinosa were obtained. Under the optimal extraction conditions,the concentration of spinosad and its extraction rate were 74.50 μg/mL and 98.18 %,respectively.
