鸡胚成纤维细胞与鸡气管黏膜上皮细胞共培养体系的建立

摘要:
【目的】 建立鸡气管黏膜上皮细胞(Chicken trachea epithelium cells,CTECs)和鸡胚成纤维细胞(Chicken embryo fibroblasts,CEFs)共培养体系,为CTECs体外培养研究与应用奠定基础。【方法】 利用细胞套皿培养方法,使CEFs与CTECs体外共培养在一个营养体系中,用含有不同促细胞生长因子(氢化可的松(HC)、转铁蛋白(TF)、人表皮生长因子(HEGF)或牛垂体提取物(BPE))和血清(胎牛血清、鸡血清)的培养液培养,观察各组CTECs的生长情况,绘制生长曲线,记录CTECs体外生长增殖的特点。【结果】 与单独培养的CTECs相比,共培养体系中的CTECs体外贴壁、生长增殖能力均显著提高,72 h能够铺满整个培养皿,并呈典型的上皮细胞“铺路石”状排列。共培养体系中的CTECs,在缺少3种重要哺乳动物气管黏膜上皮细胞培养所需的促细胞生长因子培养液中,仍能保持较好的生长能力,大大降低了CTECs的培养成本。【结论】 成功建立了CTECs与CEFs的共培养体系,为CTECs的体外培养和应用提供了技术支持。

Establishment of co-culture system of chicken embryo fibroblasts and chicken trachea epithelium cells
Abstract:
【Objective】 The purpose of this study was to lay the foundations for research and use of culture in vitro of chicken trachea epithelium cells (CTECs)by establishing co-culture system of CTECs and chicken embryo fibroblasts (CEFs).【Method】 By applying nest dish culture technology,CEFs and CTECs were co-cultured in the same system.Three kinds of culture fluids including different nutrient compositions (HC,TF,HEGF,BPE,fetal calfserum or chicken serum) were used to detect each effect on growth of CTECs.Growth curvature,proliferation and growthform of CTECs were made and observed.【Result】 We found that compared with CTECs in the single culture system,the adherence,growth and proliferation of CTECs in the co-culture system were promoted markedly and CTECs could overspread the whole culture dish after it was planted into the dish in 72 h and showed typical form “paving stone”.And the co-cultured CTECs still had good cell living with the culture fluid without three cell growth factors which are necessary for the growth of TECs of mammal.The co-culture system made culture and use of CTECs in vitro more economical.【Conclusion】 We have successfully established the technology of co culture of CTECs and CEFs,which may provide technology for CTE relative research and lay material base for basic research and application of poultry diease and at the same time save much cost for the use of CTECs in vitro.
