【目的】 考察壳聚糖絮凝法去除中药材提取液中杂质及糖类的效果,为中药材HPLC指纹图谱的建立提供理论依据。【方法】 以玄参、天麻、半夏3种糖分含量高的中药材为供试材料,分别采用醇沉法和壳聚糖絮凝法处理,考察这2种方法对中药材提取液中的糖分和有效成分的影响,比较HPLC指纹图谱的相似度。【结果】 与醇沉法相比,壳聚糖絮凝法使中药材有效成分得到了更好的保留,澄清效果明显,对糖分的去除也达到了要求;壳聚糖絮凝剂的质量浓度和加入体积分别为10 g/L和100 μL(10 mL提取液)时效果最佳;壳聚糖絮凝剂处理玄参提取液与模板的相似度为0.989 2,明显高于醇沉法(0.892 1);天麻及半夏与模板的相似度分别为0.997 3和0.979 6。【结论】 作为一种前处理方法,壳聚糖絮凝法可用于中药材HPLC指纹图谱的分析及中药材提取与精制中。 |

【Objective】 The study was done to investigate the effect of Chitosan flocculant to remove the impurities and saccharide in the extract of three Chinese medical herbs for testifying HPLC fingerprint analysis.【Method】 Three over-saccharide Chinese medical herbs(Scrophularian ningpoensis Hemsl.,Gstrodia elata Hlume and Typhonium flagelliforme Blume)were used as materials.The extract was pretreated by Chitosan or Ethanol respectively.The contents of total saccharide and active components in the extract of three Chinese medical herbs were determined,and the similarities between the extract treated by Chitosan and the extract treated by Ethanol were compared.【Result】Compared with alcohol sedimentation,Chitosan-flocculation saved more trace componets,clarification effect was obvious,and the content of total saccharide was in an equal range.Concentration and volume of chitosan-flocculation were 10 g/L and 100 μL(per 10 mL extract) respectively.Similarities of the HPLC fingerprint were analyzed,in the Scrophularian ningpoensisHemsl.,the similarity of chromatograms of samples dealt with Chitosan-flocculation and the standard fingerprint was 0.989 2,obviously higher than 0.892 1 which dealt with alcohol;the similarities of chromatographies of Gstrodia elata Hlume and Typhonium flagelliforme Blume were 0.997 3 and 0.979 6 respectively.【Conclusion】As a new pretreatment,Chitosan-flocculation could be widely used in HPLC analysis,refinement and separation of Chinese medical herbs. |
