中国部分山羊品种POU1F1基因的分子遗传特征分析

摘要:
【目的】探讨中国部分山羊品种POU1F1基因的分子遗传特征,为山羊品种鉴定提供科学资料。【方法】 以内蒙古白绒山羊(IMWC)、西农萨能奶山羊(Sa)、崂山奶山羊(LS)、关中奶山羊 (GZ)、贵州黑山羊 (GB)、马头山羊(MT)、板角山羊 (BJ)、贵州白山羊(GW)和雷州山羊 (LZ)为材料,利用分子群体遗传学方法,研究中国部分山羊品种POU1F1基因Dde I位点的分子遗传特征。【结果】 PCR扩增到450 bp的POU1F1基因(包括Exon 6和3′UTR),经限制性内切酶Dde I酶切后获得D1D1和D1D2 2种基因型,各山羊品种D1等位基因频率为0.600~1.000,D2为0.000~0.400,纯合度(Ho)为0.520~1.000,杂合度(He)为0.000~0.480,有效等位基因数(Ne)为1.000~1.923,多态信息含量(PIC)为0.000~0.365。独立χ2分析显示,POU1F1Dde I位点的基因型和等位基因分布在不同山羊品种之间存在显著(P<0.05)或极显著(P<0.01)差异。【结论】POU1F1DdeⅠ位点的基因型分布和等位基因分布与山羊品种显著相关,品种对山羊该位点的分子遗传特征有显著影响(P<0.05)。

Abstract:
【Objective】 This research analyzed the molecular genetic characteristics of goat POU1F1 gene in Chinese goat breeds,which will contribute to clarify breeds of goat.【Method】 PCR RFLP and molecular population genetics analysis were applied to study the molecular genetic characteristics of DdeI locus of POU1F1 gene in several Chinese goat breeds including Inner Mongolia white cashmere (IMWC),Xinong Saanen dairy (Sa),Laoshan dairy (LS),Guanzhong dairy (GZ),Guizhou black (GB),Matou (MT),Banjiao (BJ),Guizhou white (GW) and Leizhou (LZ).【Result】 The 450 bp PCR amplification products of goat POU1F1gene (including Exon 6 and 3′UTR) was digested by restriction enzyme DdeI and two different genotypes (D1D1 and D1D2) were found.Frequencies of allele D1,allele D2,Ho,He,Ne and PIC varied among breeds were from 0.600 to 1.000,from 0.000 to 0.400,from 0.520 to 1.000,from 0.000 to 0.480,from 1.000 to 1.923 and from 0.000 to 0.365,respectively.The chi square analysis revealed that there were significant statistical differences (P<0.05) or high significant statistical differences (P<0.01) of distributions of genotype frequencies and allele frequencies atPOU1F1 DdeI locus among analyzed goat populations.【Conclusion】 Hence,breed factor had significant effect on the molecular genetics characteristics at POU1F1 DdeI locus in goat.
