Cr6+胁迫对高丹草幼苗根质膜氧化及ATPase活性的影响

摘要:
【目的】 研究不同浓度Cr6+处理对高丹草幼苗根质膜氧化及ATPase活性的影响,阐明重金属Cr对植物的毒害机理,为铬污染地区优质牧草的种植提供理论依据。【方法】 采用不同浓度(0(对照),0.5,1.0,1.8 mmol/L) K2CrO4胁迫处理高丹草幼苗,测定根长、根干质量、根系中的Cr含量、根系O-2产生速率和H2O2、丙二醛(MDA)含量及根质膜H+-ATPase、Ca2+-ATPase活性的变化。【结果】 与对照相比,随着Cr6+处理浓度的增加,高丹草幼苗根长和根干质量均呈先升高后降低的变化趋势,O-2产生速率、H2O2和MDA含量及细胞质膜相对透性显著升高(P<0.05),且与Cr6+处理浓度呈正相关。0.5 mmol/L Cr6+浓度处理时,H+-ATPase、Ca2+-ATPase活性与对照差异不显著(P>0.05),而在1.0,1.8 mmol/L Cr6+处理时,ATPase活性显著降低(P<0.05),并且与膜脂过氧化水平呈负相关。【结论】 低浓度Cr6+处理对高丹草幼苗根系质膜ATPase活性影响较小,较高浓度Cr6+处理使ATPase活性及根生长受到抑制。

Effects of Cr6+ on lipid peroxidation and ATPase activity of Sorghum Hybrid Sudangrass (Sorghum bicolor×S.Sudanense) roots
Abstract:
【Objective】 The research investigated the effects of Cr6+ on lipid peroxidation and ATPase activity of Sorghum Hybrid Sudangrass (Sorghum bicolor×S.Sudanense) seedling roots,and explored the toxic hazard of heavy metal Cr Stress on Plant.【Method】 The effects of different concentrations of Cr6+(0(CK),0.5,1.0,1.8 mmol/L K2CrO4) on the length and dry weight of roots, generating rate of O -2,H2O2and MDA content as well as plasma membrane relative permeability,activities of H+-ATPase,Ca2+-ATPase in the plasma membrane of Sorghum Hybrid Sudangrass seedlings roots were investigated.【Result】 Compared with the control, the length and dry weight of roots increased first and then decreased with the increase of concentrations of Cr6+,which resulted in significant increase of O -2and H2O2 level and malondialdehyde(MDA)content as well as plasma membrane relative permeability in the roots(P<0.05),and there was significantly positive correlation to Cr6+ treatment concentration.Moreover,the activity of H+-ATPase and Ca2+ ATPase had no obvious change(P>0.05)under 0.5 mmol/L Cr6+ concentration,but significantly decreased with 1.0,1.8 mmol/L Cr6+ stress(P<0.05),a negative correlation was present between the level of membrance-lipid and the two enzyme.【Conclusion】 We concluded that there was unnoticeable change under low concentration of the Cr6+ in the activity of ATPase in Sorghum Hybrid Sudangrass seedlings roots plasma membrane,whereas it would dropped markedly and the root growth was repressed under relatively high Cr6+ stress.
