| 【目的】 研究秦川牛脂联素基因第2外显子的多态性及其与部分产肉性状的关系。【方法】 以405头24月龄的秦川牛为研究对象,利用PCR-SSCP和测序技术对其脂联素基因第2外显子的多态性进行检测,并对发现的多态位点与秦川牛产肉性状的相关性进行分析。【结果】 秦川牛脂联素基因第2外显子存在AA、AB和BB 3种基因型,基因型频率分别为0.696,0.207和0.097;等位基因A和B的基因频率分别为0.799和0.201。BB 基因型个体在64 bp处发生了G→C突变,导致22位的谷氨酸(GAA)转变为谷氨酰胺(CAA)。胸围、宰前活质量、胴体质量、背膘厚和眼肌面积等指标,AA型个体显著高于AB型(P<0.05);AA型个体腿臀围显著高于AB型(P<0.05),极显著高于BB型(P<0.01)。【结论】 脂联素基因第2外显子可能是影响秦川牛产肉性能的主效数量性状座位或与之紧密连锁,可作为秦川牛产肉性状的辅助选择标记。 |

| 【Objective】 The relationship between polymorphism in exon 2 of adiponectin gene and several production traits in Qinchuan beef cattle was studied.【Method】 405 Qinchuan cattle aged 24 months were selected to detect SNPs of exon2 by PCR-SSCP technique.【Result】 SSCP band is divided into 3 genotypes:AA,AB and BB;genotypes frequency are 0.696,0.207 and 0.097 respectively.The frequencies of alleles A and B are 0.799 and 0.201 respectively.The results showed that BB genetype has a loci mutation (G→C) at 64 bp.This mutation causes the change of Glu (GAA) into Gln(CAA).Statistical analysis revealed that Qinchuan cattle with AA genotype are higher than BB genotype in heart girth,slaughter weight,back fat thickness,carcass weight,loin muscle area and of AA genotype (P<0.05),and crural girth of AA genotype is significantly higher than that of AB genotype and BB genotype (P<0.01).【Conclution】 We can conclude that adiponectin gene exon2 is closely related to production traits (P<0.05),and can be used as a candidate gene for the production of high grade meat of Qinchuan beef. |
