脱氧鬼臼毒素对粘虫幼虫体壁主要成分的影响

摘要:
[目的]通过测定脱氧鬼臼毒素对粘虫(Mythimna separata Walker)幼虫体壁主要成分的影响,进一步探讨脱氧鬼臼毒素对粘虫生长发育的抑制机理.[方法]以脱氧鬼臼毒素为供试药剂,分别测定药剂对粘虫幼虫体壁表皮3种主要成分的影响,并对表皮中5类蛋白质含量变化及组分差异进行分析.[结果]脱氧鬼臼毒素对粘虫幼虫生长发育具有较强的抑制作用,且随其质量浓度的增加,粘虫幼虫的生长发育抑制率增强;在2 mg/mL剂量处理下,粘虫幼虫体壁中总脂肪相对含量显著增加,总蛋白质相对含量显著降低,几丁质含量变化不大,且各类蛋白质的比例有所改变,脱氧鬼臼毒素对水溶性非结合蛋白、氢键结合蛋白和共价键结合蛋白含量的影响大于弱键结合蛋白和电价键结合蛋白.聚丙烯酰胺凝胶电泳结果显示,处理粘虫幼虫表皮蛋白出现或缺失蛋白组分,即药剂使处理粘虫幼虫表皮蛋白生成和消解回吸滞后.[结论]脱氧鬼臼毒素对粘虫幼虫体壁主要成分及蛋白代谢有较大影响.

Influence of Deoxypodophyllotoxin on intugement components of Mythimna separate Walker
Abstract:
【Objective】 The study detected the influence of Deoxypodophyllotoxin on intugement components of Mythimna separata Walker,to probe Deoxypodophyllotoxin growth inhibition mechanism to larval of M.separata.【Method】 The influence of Deoxypodophyllotoxin on the intugement components and five protein contents of Mythimna separata Walker were determined and analyzed.【Result】 Deoxypodophyllotoxin had a great insect growth inhibition function to the larvae.The research showed that it increased the contents of fats,reduced the contents of cuticle proteins,while the contents of chitin was not significantly different from that of CK.At the same time,it also changed the ratios of various cuticle proteins.Deoxypodophyllotoxin can reduce the contents of several cuticle proteins,especially that of water-soluble proteins,Hydrogen-bond conbined proteins and Covalent-bond conbined proteins.Gel electrophoresis studies showed that protein constituents of Deoxypodophyllotoxin-treated larvaes were different from CK.Some protein bands appeared and disappeared later in Deoxypodophyllotoxin-treated larvaes than CK.【Conclusion】 Deoxypodophyllotoxin has a great effect on cuticle components and protein metabolization of Mythimna separata.
