猪繁殖与呼吸综合征病毒变异株的致病性试验

摘要:
[目的]研究从江西、湖南发生持续高热的病猪体内分离的4株NSP2基因缺失的猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)Jiangxi-3、Jiangxi-4、Hunan-1、Hunan-2的致病力.[方法]通过Reed-Muench法,测定4株PRRSV(第3代)的TCID50,将毒株分别接种PRRSV和猪圆环病毒2型(PCV2)抗原与抗体均为阴性的健康猪,各设同居试验猪,通过检测体温、临床症状观察、RT-PCR、ELISA、病理切片等方法检测病毒的致病性.[结果]4株病毒的TCID50为10-3.75~10-4.5/mL.攻毒试验猪和同居试验猪均表现典型猪繁殖与呼吸综合征临床症状,最后试验猪均死亡.病理切片也显示典型的猪繁殖与呼吸综合征病理变化,并在脑组织内检测到PRRSV核酸.[结论]4株PRRSV分离株的致病力增强.

Pathogenicity test of PRRSV variation
Abstract:
【Objective】 The pathogenicity of PRRSV variations(Jiangxi-3,Jiangxi-4,Hunan-1,Hunan-2) isolated from pigs of the so-called “Mystery Fever Disease Syndrome” in pig herds in China were researched.【Method】 TCID50 of the four PRRSV isolates were detected by Reed-Muench-methond,The healthy pigs which were negative of antibody and antigen of PCV2 and PRRSV were divided into 4 groups.One group was challenged by 4 isolations respectively,another was sentinel pig.The pigs were isolated artificial feeding.Pathogenicity was evaluated by temperature detection,clinical symptom,RT-PCR,ELISA and pathological section.【Result】 The TCID50 of the isolations were 10-3.75-10-4.5/mL respectively.Typical PRRS symptom appered on the challenged and sentinel pigs.It is noticeable that all pigs showed symptoms of different levels of diarrhea,and sometimes with nerve.The results of pathological showed typical pathological change of PRRSV,and PRRSV RNA had been detected. 【Conclusion】 The result showed the pathogenicity of 4 PRRSV variation isolates were intensified.
