西北粳稻成熟胚高频再生体系研究

摘要:
[目的]研究基因型、培养基、激素、碳源的种类和质量浓度对西北粳稻成熟胚培养特性的影响,建立西北粳稻成熟胚离体再生体系。[方法] 以4个西北粳稻品种宁粳16、宁粳34、宁粳35和宁粳38的成熟胚为材料,利用组织培养法,对水稻成熟胚进行脱分化和再分化,获得再生植株,研究培养基、基因型及激素、碳源质量浓度和种类对西北粳稻成熟胚愈伤组织诱导和分化的影响。[结果] 基因型对西北粳稻成熟胚培养的影响较大,参试材料中宁粳34的培养特性最好;N6和NB 2种培养基对西北粳稻培养效果的影响随粳稻品种基因型的不同而不同;诱导培养基中添加2,4-D的质量浓度以2 mg/L为好,添加0。2~0。5 mg/L的6-BA可提高西北粳稻成熟胚的愈伤组织诱导率和绿苗分化率。以麦芽糖和蔗糖为碳源时,西北粳稻愈伤组织诱导率和绿苗分化率均较高,蔗糖质量浓度为30~40 g/L时培养效果最佳。[结论] 基本明确了基因型、培养基、激素、碳源对西北粳稻成熟胚离体培养特性的影响效果,初步建立了西北粳稻成熟胚高频再生体系。

Research on high frequency regenerated system in mature embryo of northwest japonica rice of China
Abstract:
【Objective】 The effects of mature embryo culture were studied including influence of genotype,culture medium,hormone,carbon sources and its concentration with northwest japonica rice cultivars to establish vitro regeneration system.【Method】 Using mature embryos of four northwest japonica rice Ningjing 16,Ningjing 34,Ningjing 35 and Ningjing 38,and utilizing the plant tissue culture method,the mature embryo of rice was dedifferentiated and differentiated again, then the rice was regenerated.The effects of callus induction and green plantlet differentiation were studied including influence of genotype,culture medium,hormone,carbon sources and concentration.【Result】 Genotype was ranked at the top of the factors,and Ningjing 34 had high culture ability.Using N6 culture medium and NB culture medium,different genotypes had different culture abilities on mature embryo culture. The addition of 2 mg/L 2,4-D to induction medium was favorable for testing cultivars.In induction culture medium,adding 0.2-0.5 mg/L 6-BA can improve mature embryo culture frequency.Sucrose and maltose were all good carbon source to induce and differentiate callus.And 30-40 g/L was best sucrose concentration for callus inducing and differentiation.【Conclusion】 Using mature embryo of northwest japonica rice,the culture effect of genotype,culture medium,hormone,carbon source was defined basically,and regeneration system was established preliminarily.
