[目的]明确陕西省生产苹果汁中脂环酸芽孢杆菌(Alicyclobacillus)的分布情况.[方法]对健康苹果苹果汁和软腐苹果腐烂部位中的嗜酸耐热菌进行了分离、纯化,并以1株酸土脂环酸芽孢杆菌(A.acidoterrestris)DSM3922为标准菌株,对分离菌株进行了个体形态、菌落形态观察和嗜酸性、耐热性、生理生化特性试验,最后用表型聚类法分析了供试菌株的亲缘关系.[结果]分别从苹果汁、腐烂苹果中分离到50和5个疑似菌落,嗜酸耐热特性试验表明,其中有14株属嗜酸耐热菌,且均是芽孢杆菌,大小为2.0~5.0 μm;其菌落表面湿润,中心突起,有同心圆,半透明,不易挑起,形状圆形,直径为1.5~5 mm;接触酶、脲酶检验结果均呈阳性,都可利用肌醇、甘油、L-山梨糖为惟一碳源,且利用葡萄糖均不能产酸产气.鉴定结果确定分离菌株均属于脂环酸芽孢杆菌属的不同种,其中TAB-3号菌与标准菌株最为接近.[结论]陕西省苹果汁中的耐热菌,除酸土脂环酸芽孢杆菌以外,还有脂环酸芽孢杆菌属的其他菌,其主要分属于A.acidoterrestris、A.acidocaldarius、A.pomorum 3个种. |

Isolation and identification of Alicyclobacillus from Shaanxi apple juice |

Abstract: |
【Objective】 The study ascertained the distribution and type of Alicyclobacillus sp in apple juice in Shaanxi.【Method】 Alicyclobacillus sp from apple juice and rotten apples was isolated and purified.Using Alicyclobacillus acidoterrestris as a standard strain,the isolated strains were researched by morphological,biochemical and physiological methods in this paper.Cluster analysis of morphological characters was used to rearch relationship between tested strains.【Result】 50 isolates and 5 isolates were obtained from apple juice and rotten apple respectively,and 14 strains fell into acidophilic and heat-resistant bacteria:all rod and produce spores,2.0-5.0 μm,round clony,semitransparent and uneasy priching,moist and convex clony surface,1.5-5 mm in diameter.Urease,catalase and nitrate induction all show positive.All tested strains could grow well with only glurose,glycerol and L-sorbose as carbon source.No gas and acid could be produced by using glurose.All isolated strains belonged to Alicyclobacillus,of which TAB-3 was mostly related to standard strain.【Conclusion】 In addition to Alicyclobacillus acidoterrestris,heat-resistant bacteria in Shaanxi apple juice include other strains of Alicyclobacillus,which are the three strains of A.acidoterrestris,A.acidocaldarius and A.pomorum. |
